摘 要:目的建立一种高效快速的超高效液相色谱串联四极杆轨道阱质谱(UHPLC-Q-Exactive Orbitrap MS)法,对退黄合剂的化学成分进行表征。方法采用Waters ACQUITY UPLC BEH C_(18)色谱柱(100 mm×2.1 mm,1.7μm),以流动相0.1%甲酸水溶液(A)-乙腈(B)进行梯度洗脱,流速为0.3 mL/min,进样量为2μL,柱温为40°C。质谱采用电喷雾离子源(ESI)负离子模式,质谱扫描范围为m/z 100~1500。结果通过对照品、数据库及参考文献共鉴定出退黄合剂中77种化学成分,其中包括黄酮类30种、多酚类18种、有机酸10种、萜类9种、蒽醌类9种、其他类1种。结论所建立的方法可高效、快速地鉴定退黄合剂的化学成分。Objective An effective and rapid analysis method by UHPLC-Q-Exactive Orbitrap MS was established to identify the components of Tuihuang Mixture.Methods The analysis was performed on a Waters ACQUITY UPLC BEH C_(18)(100 mm×2.1 mm,1.7μm)HPLC column.The mobile phase consisted of 0.1%formic acid(A)and acetonitrile(B)was used as gradient elute.The flow rate was 0.3 mL·min–1,column temperature was 40℃,and injection volume was 2μL.The ESI was operated in negative mode with the scanning range of m/z 100~1500.Results A total of 77 chemical components were identified by standards,database and references,including 30 flavonoid compounds,18 polyphenol compounds,10 organic acid compounds,9 terpenoids compounds,9 anthraquinone compounds,1 other constituent.Conclusion The established UHPLC-Q-Exactive Orbitrap MS technology can rapidly analyze and identify the components of Tuihuang Mixture.
