摘 要:蛋白邻近标记(proximity labeling,PL)技术是指通过融合特定的催化酶,实现对目标蛋白相互作用蛋白的原位标记,再采用质谱或测序分析鉴定标记蛋白。该技术能够在瞬时动态条件下精确识别蛋白质相互作用,是传统分析手段的重要补充,在蛋白质相互作用研究中具有广阔应用前景。本文介绍了各类PL酶的工作原理、分类及优缺点;论述了该技术在研究蛋白质与蛋白质相互作用中的应用;讨论了PL技术的优势、目前存在的问题和发展趋势,以期为该技术的应用提供参考。Proximity labeling(PL)technology refers to the in-situ labeling of protein-interacting proteins of interest by fusing specific catalytic enzymes,followed by mass spectrometry or sequencing analysis to identify the tagged proteins.This technique can accurately identify protein-protein interactions under transient dynamic condition,which is an important supplement to traditional analytical methods,and has a broad application prospect in the study of protein-protein interactions.In this paper,the working principle,classification,advantages and disadvantages of various PL enzymes were introduced,then the application of this technology in the study of protein-protein interactions was discussed,and finally the advantages,existing problems and development trends of adjacent labeling technology were discussed,in order to provide reference for the application of this technology.
