摘　　要：目的建立脂肪间充质干细胞(Adipose Derived Stem Cells,ADSCs)与K562细胞共培养体系,探讨脂肪间充质干细胞对K562细胞增殖的影响。方法原代分离培养ADSCs至第3代并鉴定。ADSCs和K562细胞共培养,以单独K562细胞孔为对照,均分别接种24孔。于第1、3、5、7 d分别选择6孔细胞,计数细胞并绘制细胞生长曲线。运用广义估计方程对重复测量资料进行统计分析。结果分离培养出ADSCs并鉴定成功。细胞生长曲线图显示,随着培养时间从第0 d至第7 d,单独培养组的K562细胞和共培养组的K562细胞增殖速度均越来越快,培养至第3、5、7 d时,单独培养组的K562细胞平均数量分别为(2.85±0.08)×10^(4)/mL、(10.70±0.36)×10^(4)/mL、(39.00±0.30)×10^(4)/mL,共培养组的K562细胞平均数量分别为(2.67±0.11)×10^(4)/mL、(10.01±0.19)×10^(4)/mL、(38.00±0.37)×10^(4)/mL,2组细胞同一天的增殖细胞数量比较,差异均有统计学意义(P均<0.001)。结论ADSCs对K562细胞的增殖有抑制作用。Objective To establish a co-culture system of adipose-derived stem cells(ADSCs)and K562 cells,and to explore the effect of adipose mesenchymal stem cells on the proliferation of K562 cells.Methods Primary isolation of ADSCs was cultured to the 3rd generation and identified.ADSCs and K562 cells were co-cultured,K562 cell wells were used as control alone,and 24 wells were seeded respectively.Six-well cells were selected on days the 1st,3rd,5th,and 7th,and cells were counted and cell growth curves were plotted.The generalized estimation equation was used to statistically analyze the repeated measurement data.Results ADSCs were isolated and successfully identified.The cell growth curve showed that with the culture time from the 0th day to the 7th day,the proliferation rate of K562 cells in the single culture group and the co-culture group was getting faster and faster,and the average number of K562 cells in the single culture group was(2.85±0.08)×10^(4)/mL,(10.70±0.36)×10^(4)/mL,and(39.00±0.30)×10^(4)/mL,the average number of K562 cells in the co-culture group were(2.67±0.11)×10^(4)/mL,(10.01±0.19)×10^(4)/mL,and(38.00±0.37)×10^(4)/mL,respectively,and the differences in the number of proliferating cells between the two groups on the same day were statistically significant(all P<0.001).Conclusion ADSCs have an inhibitory effect on the proliferation of K562 cells.
