摘 要:目的 探讨加味逍遥散是否通过外泌体对神经细胞突起的发育发挥调节影响作用。方法 采用体外神经细胞、星形胶质细胞非接触式共培养系统模拟体内中枢系统神经细胞和星形胶质细胞相互交错,相互影响的联系,后加入肝癌细胞来源外泌体、经加味逍遥散含药血清干预后的肝癌细胞来源外泌体,通过比较二者对神经细胞-星形胶质细胞共培养系统中神经细胞树突的长度、分支,兴奋性突触及相关突触蛋白的数量等的影响。结果 经肝癌细胞来源外泌体干预的神经细胞树突的长度、分支,兴奋性突触及相关突触蛋白的数量均较空白组减少,而经过加味逍遥散含药血清干预后的肝癌细胞外泌体可显著增加共培养系统中神经细胞的树突长度、分支,兴奋性突触及相关突触蛋白的数量。结论 加味逍遥散对肝癌来源外泌体造成的神经细胞突起的异常损伤具有修复和保护作用,即加味逍遥散可能通过外泌体途径干预和修复受损的神经细胞。Objective Exploring whether Jiawei Xiaoyao San(JWXYS) has a regulatory effects on the growth of neural cell protrusions through exosomes. Methods The non-contact co culture system of neural cells and astrocyte in vitro was used to simulate the interleaving and influencing relationship between cells in the central nervous system in vivo, then added the exosomes separately derived from hepatoma cells and hepatoma cells dealt with JWXYS containing serum. Finally, compare the effects of the two on the length and branch of nerve cell dendrites, and the number of excitatory synapses and sunaptic associated proteines, etc in the neurocyte Astrocyte co culture system. Results Compared with the blank group, the length and branch of the nerve cell dendrites, as well as the number of excitatory synapses and sunaptic associated proteines intervened by exosomes derived from hepatoma cells were decreased. However, the exosomes derived from hepatoma cells which were dealt with JWXYS containing serum can significantly increase the length and branch of the nerve cell dendrites, and the number of excitatory synapses and sunaptic associated proteines in the co-culture system. Conclusion JWXYS could repair and protect the abnormal damage of neural cell protrusions caused by the exosomes derived from hepatoma cells. In other words, JWXYS may intervene and repair damaged nerve cells through the exosomes pathway.
