摘  要:本研究旨在评价不同免疫刺激剂的安全性及免疫增强特性,为后续亚单位疫苗的制备提供佐剂选择依据。本研究利用不同免疫刺激剂皮下免疫小鼠,免疫两次,间隔1周,最后1次免疫后的第7天处死小鼠,观察小鼠脏器大体病变;利用ELISA方法检测小鼠血清中细胞因子的变化;取注射部位及主要脏器进行组织病理学观察。研究结果表明,免疫刺激剂壳聚糖和HMT13均引起促炎细胞因子IFN-γ显著增加,HMT13和ISA201会引起大量Ig G的产生;此外,壳聚糖刺激还可引起大量IL-17的分泌。本研究通过观察小鼠主要脏器及注射部位的大体和组织病理学变化以及检测小鼠血清中细胞因子的变化等方法评价了免疫刺激剂Alum、壳聚糖、α-半乳糖基神经酰胺、HMT13、ISA201的安全性及免疫增强特性,为后续亚单位疫苗佐剂的选择提供了数据支持及参考依据。The aim of this study was to evaluate the safety and immune-enhancing properties of dif-ferent immunostimulants,and to provide a basis for the selection of adjuvants for the subsequent preparation of subunit vaccines.Mice were immunized subcutaneously with different immune stimulants and immunized twice,with an interval of 1 week.Mice were executed on the 7th day after the last immu-nization,and the gross lesions of the organs of mice were observed.Serum was collected and the changes of cytokines in serum were detected by ELISA.The injection sites and major organs were taken for histopathological observation.The results showed that chitosan and HMT13 caused a significant in-crease in the pro-inflammatory cytokine IFN-,and HMT13 and ISA201 caused a large amount of IgG pro-duction.In addition,chitosan stimulation also caused a large amount of IL-17 secretion.In the study,the safety and immune-enhancing properties of the immunostimulants Alum,chitosan,α-galactosylceramide,HMT13,and ISA20l were evaluated by observing the gross and histopathological changes in the major organs and injection sites of mice and detecting the changes of cytokines in the serum of mice.These findings provide data support and a reference basis for the selection of adjuvants for subsequent subunitvaccines.
