摘 要:为了探究葛根素对镉(Cd)致大鼠大脑皮质线粒体损伤的保护效应,将24只SD大鼠随机分为对照组(Con)、葛根素组(Pur)、镉组(Cd)、镉与葛根素共处理组(Cd+Pur)。Pur组、Cd+Pur组大鼠每日以每千克体重200 mg葛根素(溶解于羧甲基纤维素钠)灌胃,Con组、Cd组大鼠每日用相同剂量羧甲基纤维素钠灌胃,持续7周。从第4周开始,Cd组、Cd+Pur组大鼠在灌胃的同时自由饮用75 mg/L镉水溶液,持续4周。试验结束后,剖杀采集大鼠大脑皮质,火焰原子吸收光谱法检测镉含量,化学发光法检测ATP水平,比色法检测NAD^(+)/NADH比率、Mn SOD活性和H_(2)O_(2)水平,实时荧光定量PCR法检测线粒体氧化应激相关基因(GST、CAT、Gpx、UCP1、UCP2和UCP3)转录水平。结果显示,与Con组相比,Cd组大鼠大脑皮质镉含量、H_(2)O_(2)水平、线粒体氧化应激相关基因转录水平显著上升(P<0.05),ATP水平和NAD^(+)/NADH比率、MnSOD活性显著下降(P<0.05);与Cd组相比,Cd+Pur组大鼠大脑皮质镉含量、H_(2)O_(2)水平和线粒体氧化应激相关基因转录水平显著下降(P>0.05),MnSOD活性显著上升(P<0.05),ATP水平和NAD^(+)/NADH比率上升但无显著性差异(P>0.05)。结果表明,葛根素可降低镉暴露大鼠大脑皮质镉含量,同时葛根素通过缓解线粒体功能障碍、线粒体氧化应激减轻了镉致大鼠大脑皮质线粒体损伤。To study the protective effect of puerarin on cadmium-induced mitochondrial damage,twenty-four SD rats were randomly divided into control group(Con),cadmium group(Cd),puerarin group(Pur),cadmium and puerarin co-treated group(Pur+Cd).The Pur group and Pur+Cd treated group underwent gavage of puerarin dissolved in sodium carboxymethyl cellulose at a dose of 200 mg/kg body weight,while the Con group and Cd treated group were given the same dose of sodium carboxymethyl cellulose by gavage every day for 7 weeks.The Cd group and Pur+Cd treated group received drinking water containing 75 mg/L Cd plus the gavage from the 4th week and lasted for 4 weeks.After the experimental trail comple-tion,the rats were dissected and collected the cerebral cortices.The cadmium contents were detected by flame atomic absorption spectrometry.ATP level was detected by luminometer.The ratio of NAD^(+)/NADH,the activity of MnSOD and the level of H_(2)O_(2)were detected by colorimetry.The transcriptional levels of mitochondrial oxidative stress-related genes were detected by real-time quantitative polymerase chain reaction.The results showed that compared with the Con group,the cadmium contents,the level of H_(2)O_(2)and the transcription levels of mitochondrial oxidative stress-related genes significantly increased(P<0.05),while ATP level,the ratio of NAD^(+)/NADH and the activity of MnSOD significantly decreased(P<0.05)in cerebral cortices of rats in Cd group.Compared with the Cd group,the cadmium contents,the level of HO2 and the transcription levels of mitochondrial oxidative stress-related genes significantly decreased(P<0.05),while the activity of MnSOD significantly increased(P<0.05),ATP level and the ratio of NAD^(+)/NADH increased without significant differences(P>0.05)in cerebral cortices of rats in Cd+Pur group.The results indicate that puerarin reduces cadmium contents in the cerebral cortices of cadmium-exposed rats.Puerarin can alleviate the mitochondrial damage caused by cadmium in the rat cerebral cortices by alleviating mito
