牛肉中禁用药物高通量筛查技术研究与应用

摘  要:文章结合液相色谱高分辨飞行时间质谱建立高效快速检测牛肉中17种禁用药物高通量筛查检测技术。样品用0.2%甲酸(80:20)的乙腈水溶液提取,KNORTH~?Clean-up LPAS(Lipid and protein adsorbent)净化柱净化。Phenomenex Kinetex F5色谱柱分离,以0.1%甲酸水和0.1%甲酸乙腈作为正离子扫描流动相,水和乙腈作为负离子扫描流动相,梯度洗脱,流速为0.3 mL/min,柱温为40℃,质谱采用电喷雾离子源,分为正离子和负离子2种扫描模式进行扫描。结果显示,17种禁用药物在牛肉中的检出限和定量限分别为0.04~0.28μg/kg和0.12~0.95μg/kg,回收率为71.9%~115.2%。17种禁用药物在5~200μg/mL范围内线性关系良好,相关系数(R2)大于0.98。该研究所建立的高通量筛查技术适用于牛肉中17种禁用药物的检测。该方法快捷简单,能够有效检测出牛肉中的禁用药物成分,具有较高的准确性和可靠性。The article combines liquid chromatography-high-resolution time-of-flight mass spectrometry to establish an efficient and rapid high-throughput screening and detection technology for 17 prohibited drugs in beef.The samples were extracted with 0.2%formic acid(80:20)acetonitrile water solution,purified using KNORTH~?Clean up LPAS(Lipid and protein adsorbent)column.Separation was achieved on a Phenomenex Kinetex F5 column,0.1%formic acid water and 0.1%formic acid acetonitrile used as positive ion scanning mobile phase,water and acetonitrile used as negative ion scanning mobile phase,gradient elution,flow rate of 0.3 mL/min,column temperature of 40℃,electrosprayion ionization source is used for mass spectrometry,which is divided into positive ion and negative ion scanning modes for scanning.The results showed that the detection limits and quantification limits of 17 prohibited drugs in beef were 0.04–0.28μg/kg and 0.12–0.95μg/kg,with recovery rate between 71.9%to 115.2%.The linear relationships for the 17 prohibited drugs were excellent within the range of 5μg/mL to 200μg/mL,and the correlation coefficient(R~2)greater than 0.98.The high-throughput screening technology established by in this research is suitable for the detection of 17 prohibited drugs in beef.This method is fast and simple,and can effectively detect prohibited drug components in beef with high accuracy and reliability.
