摘  要:以马尾藻为原料,探讨岩藻黄素的提取工艺,并对纯化后的岩藻黄素进行体外抗氧化和抗炎作用研究。结果表明,马尾藻岩藻黄素提取液在4℃避光保存25 d很稳定;通过单因素试验和正交试验确定最佳提取条件:以新鲜冷冻马尾藻为原料,提取溶剂为90%乙醇∶丙酮=3∶1(体积比),料液比为1∶15(g/mL),提取温度为50℃、100 r/min振荡提取6 h后再超声辅助提取15 min,在此条件下岩藻黄素提取率为0.816 mg/g海藻湿重。抗氧化试验结果显示,马尾藻岩藻黄素的氧化自由基吸收能力(oxygenradicalabsorbancecapacity,ORAC)值为1761μmolTE/g,对DPPH自由基和羟自由基的清除率IC50分别为0.102 mg/mL和0.146 mg/mL。细胞实验结果表明,100μg/mL以下的马尾藻岩藻黄素对小鼠小胶质瘤细胞(BV⁃2)无明显毒性;与脂多糖(lipopolysaccharide,LPS)诱导的模型组相比,60μg/mL的马尾藻岩藻黄素显著降低细胞上清中一氧化氮(nitricoxide,NO)和白细胞介素⁃6(interleukin-6,IL-6)含量。综上所述,马尾藻中岩藻黄素的提取率与很多因素密切相关,并具有显著的抗氧化和抗炎作用。The extraction conditions of fucoxanthin from Sargassum were optimized and the antioxidant and anti⁃inflammatory effects of the purified fucoxanthin were studied in vitro.The results showed that the fucoxan⁃thin extract of Sargassum was stable after storage at 4℃in the dark for 25 d.The extraction conditions were op⁃timized by single factor and orthogonal tests as follows:frozen fresh Sargassum as the raw material,90%etha⁃nol mixed with acetone at a volume ratio of 3∶1 as the solvent,solid⁃to⁃liquid ratio of 15∶1(g/mL),extraction at 50℃and 100 r/min for 6 h,followed by ultrasound⁃assisted extraction for 15 min.The yield of fucoxanthin reached 0.816 mg/g(wet weight)under these conditions.The extracted fucoxanthin showed the oxygen radical absorbance capacity(ORAC)of 1761μmol TE/g and the IC50 values of 0.102 mg/mL and 0.146 mg/mL for scavenging DPPH free radicals and hydroxyl free radicals,respectively.The results of cell experiments showed that extracted fucoxanthin with the concentration below 100μg/mL had no significant toxicity to mouse small glioma cells(BV⁃2 cells).Compared with the model group induced by lipopolysaccharide(LPS),60μg/mL Sargassum fucoxanthin significantly lowered the nitric oxide(NO)and interleukin⁃6(IL⁃6)levels in the cell supernatant.In conclusion,the yield of fucoxanthin extracted from Sargassum was associated with multiple fac⁃tors,and the extracted fucoxanthin exhibited significant antioxidant and anti⁃inflammatory effects.
