软肝逐瘀方对高脂饮食诱导的小鼠非酒精性脂肪性肝炎的疗效评价

摘　　要：目的观察软肝逐瘀方治疗高脂饮食诱导的小鼠非酒精性脂肪性肝炎(nonalcoholic steatohepatitis,NASH)的疗效并评价抗炎活性,为其临床应用提供数据支持。方法将42只C57BL/6小鼠随机分为模型组、空白组、水飞蓟宾组、软肝逐瘀方低、中及高剂量组共6组,每组7只。用高脂饲料喂养14周诱导小鼠NASH模型,造模成功后软肝逐瘀方各剂量组和水飞蓟宾组在高脂饲料喂养基础上每日每只小鼠1 ml药液灌胃给药13周,记录每组小鼠体质量变化。停止给药2周后处死,取血检测血清谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、层黏蛋白(laminin,LN)、透明质酸(hyaluronic acid,HA)、白介素(interleukin,IL)-2、IL-4、IL-6、IL-10及α干扰素(interferonα,IFN-α)水平。进行苏木精-伊红(hematoxylin-eosin,HE)、油红O脂肪及Masson染色观察肝脏脂肪变性、脂滴堆积及肝纤维化情况。结果与模型组相比,软肝逐瘀方组小鼠体质量显著下降(P<0.05),软肝逐瘀方高剂量组血清AST水平显著下降(P<0.05),软肝逐瘀方中、高剂量组血清ALT水平显著下降(P<0.05),IL-4、IL-6、IL-10、IFN-α水平显著下降(P<0.05),软肝逐瘀方组的HA水平显著下降(P<0.05)。HE、油红O及Masson染色结果显示,软肝逐瘀方中、高剂量组小鼠的肝纤维化和肝脏炎症显著减轻。结论软肝逐瘀方能够有效降低高脂饮食诱导的小鼠体质量,并且改善小鼠肝功能和肝纤维化。Objective To observe the efficacy and anti-inflammatory activity of Ruangan Zhuyu formula in the treatment of high-fat diet-induced nonalcoholic steatohepatitis(NASH)in mice,and to provide data support for its clinical application.Methods Forty-two C57BL/6 mice were randomly divided into model group,normal group,Silibinin Capsules group,Ruangan Zhuyu formula low,medium,and high dosage groups,seven mice in each group.The NASH model was induced in mice by feeding with high-fat diet for 14 weeks.After successful modelling,Ruangan Zhuyu formula each dose groups and the Silibinin Capsules group were given 1 ml drug solution every day by intragastric administration for 13 weeks on the basis of high-fat diet.The changes of body mass in each group were recorded.The mice were executed 2 weeks after the administration was stopped,and blood samples were collected to detect the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),laminin(LN),hyaluronic acid(HA),interleukin(IL)-2,IL-4,IL-6,IL-10 and interferonα(IFN-α).Hematoxylin-eosin(HE),oil-red,and Masson staining were used to observe the hepatic steatosis,fat droplet accumulation,and hepatic fibrosis.Results Compared with the model group,the body mass of mice in Ruangan Zhuyu formula group decreased significantly(P<0.05).The serum AST level in Ruangan Zhuyu formula high-dose group decreased significantly(P<0.05),the serum ALT level in Ruangan Zhuyu formula medium and high-dose groups decreased significantly(P<0.05),the level of IL-4,IL-6,IL-10 and IFN-αdecreased significantly(P<0.05).The level of HA in Ruangan Zhuyu formula group decreased significantly(P<0.05).The results of HE,oil red and Masson staining showed that liver fibrosis and liver inflammation were significantly reduced in mice in Ruangan Zhuyu formula medium and high-dose groups.Conclusion Ruangan Zhuyu formula can effectively reduce the body mass of mice induced by high-fat diet,and improve the liver function and liver fibrosis of mice.
