摘 要:目的基于网络药理学探讨白术、枳实改善胰岛素抵抗(IR)的分子机制。方法从中药数据库获取白术、枳实的主要活性成分及靶点,通过疾病数据库获取IR靶点,取交集。对二者交集靶点进行蛋白质互作(PPI)分析,构筑PPI网络;进行富集分析,构建“成分-IR靶点-通路”网络;最后进行分子对接验证。结果白术-枳实改善IR的主要活性成分为木犀草素、川陈皮素、柚皮素等,核心靶点有肿瘤坏死因子(TNF)、肿瘤蛋白P53(TP53)、蛋白激酶B(AKT1)、白细胞介素-6(IL-6)等;分子对接验证显示,大部分核心靶点与主要活性成分的结合活性较好。白术、枳实改善IR的生物学过程包括激素应答、细胞对脂质的反应、调节炎症反应等,作用于内分泌抵抗、HIF-1、AMPK等信号通路,功能主要为调节糖脂代谢等。结论白术、枳实可能通过木犀草素、川陈皮素等活性物质对AMPK、NF-κB等信号通路发挥调节作用,初步揭示了二者多成分、多靶点、多通路改善IR的作用机制。Objective To explore the molecular mechanism of the improvement of insulin resistance(IR)by Atractylodes macrocephala Koidz-Aurantii Fructus Immaturus on the basis of network pharmacology.Methods The main active components and their targets were obtained from traditional Chinese medicine database,and the IR targets were obtained from disease database.The protein interaction between the two intersecting targets was analyzed to construct PPI network.The potential IR targets of Atractylodes macrocephala Koidz-Aurantii Fructus Immaturus were enriched and analyzed,and a“component-IR target-pathway”network was constructed.Finally,the molecular docking was verified.Results The main active components of the IR improvement were luteolin,tangerine,naringin,etc.The core targets were TNF,TP53,AKT1,IL-6,etc.Molecular docking verification showed that most of the core targets had good binding activity with the main active components.The biological process of improving IR includes hormone response,cell response to lipids,regulation of inflammation,etc.It acts on endocrine resistance,HIF-1,AMPK and other signaling pathways,and its function is mainly to regulate glucose and lipid metabolism.Conclusion Atractylodes macrocephala Koidz-Aurantii Fructus Immaturus may regulate AMPK,NF-κB and other signaling pathways through active substances such as luteolin and tangerine,which initially revealed the mechanism of multi-component,multi-target and multi-pathway improvement of IR.
