摘 要:目的探讨丹参酮ⅡA对斑马鱼尾鳍创面修复的作用及其可能机制。方法取48条野生型或转基因斑马鱼切尾构建尾鳍创伤模型后,随机均分为模型组及低浓度丹参酮ⅡA(50μg/mL)、中浓度丹参酮ⅡA(100μg/mL)和高浓度丹参酮ⅡA(200μg/mL)组;另取未切尾的12条野生型或转基因斑马鱼作为对照组。培养3 d后记录野生型斑马鱼尾鳍再生长度,观察转基因斑马鱼再生尾鳍中性粒细胞数量,并分析丹参酮ⅡA对野生型斑马鱼再生尾鳍中活性氧(ROS)含量、细胞凋亡及TNF-α和IL-6 mRNA表达的影响。结果模型组尾鳍再生长度、中性粒细胞数量、ROS含量、细胞凋亡数以及TNF-α和IL-6 mRNA表达均较对照组增加(P<0.05);与模型组相比,低浓度丹参酮ⅡA组、中浓度丹参酮ⅡA组、高浓度丹参酮ⅡA组尾鳍再生长度呈浓度依赖性地增加,而中性粒细胞数量、ROS含量、细胞凋亡数以及TNF-α和IL-6 mRNA表达呈浓度依赖性地减少(P<0.05)。结论丹参酮ⅡA可清除炎症细胞,抑制炎症因子分泌,从而促进斑马鱼尾鳍创面修复。Objective To investigate the effect and underlying mechanism of tanshinoneⅡA on the wound repair of zebrafish caudal fin.Methods The tails of 48 wild or transgenic zebrafishes were cut for establishing the caudal fin wound model,which then were equally divided into four groups of A(model control),B1(treated with tanshinoneⅡA 50μg/mL),B2(treated with tanshinoneⅡA 100μg/mL)and B3(treated with tanshinoneⅡA 200μg/mL).Another 12 wild or transgenic zebrafishes without tail cutting were taken as the blank controls(group C).The regeneration length of the caudal fin was recorded after culture for 3 days.The number of neutrophils in the regenerated zebrafish caudal fin was observed.The effect of tanshinoneⅡA on the content of ROS,cell apoptosis and mRNA expressions of TNF-αand IL-6 in the regenerated zebrafish caudal fin were analyzed.ResultsThe regeneration length of caudal fin,number of neutrophils,content of ROS,number of cell apoptosis and mRNA expressions of TNF-αand IL-6 in group A were higher than those in group C(P<0.05).Compared with group A,the regeneration length of caudal fin was increased,while the number of neutrophils,content of ROS,number of cell apoptosis and mRNA expressions of TNF-αand IL-6 were decreased in a dose-dependent manner in groups of B1,B2 and B3(P<0.05).Conclusion TanshinoneⅡA can clear the inflammatory cells,inhibit the secretion of inflammatory factors,and promote the wound repair of zebrafish caudal fin.
