摘 要:目的研究壮药清毒伸筋颗粒对佐剂性关节炎(adjuvant arthritis,AA)大鼠的干预作用。方法采用弗氏完全佐剂诱导AA大鼠模型,随机分为模型对照组、阳性对照组(万通筋骨片,0.15 g·kg^(–1))、清毒伸筋颗粒低、中、高剂量组(3.9、7.8、15.6 g·kg^(–1)),每组10只,另设10只正常大鼠为正常对照组。各组按相应药物剂量灌胃给药,每日1次,连续30 d。定期检测大鼠痛阈值、足肿胀度、关节炎指数评分;给药结束后,采用酶联免疫吸附(ELISA)检测血清免疫球蛋白G(IgG)、免疫球蛋白M(IgM)、肿瘤坏死因子-α(TNF-α)及白细胞介素1β(IL-1β)水平;苏木素-伊红(HE)染色观察踝关节病理变化并进行评分。结果与正常对照组比较,AA大鼠的痛阈显著降低,足肿胀度、关节炎指数、血清炎症细胞因子水平显著升高,踝关节滑膜层、软骨及骨质层损伤严重,踝关节病理评分显著升高(P<0.05,P<0.01);与模型组比较,清毒伸筋颗粒可显著提高AA大鼠痛阈,降低足肿胀度、关节炎指数、血清炎症细胞因子水平,减轻踝关节滑膜层、软骨及骨质层损伤,降低踝关节病理组织评分(P<0.05,P<0.01)。结论壮药清毒伸筋颗粒对AA大鼠具有治疗作用,其机制可能与减少炎症因子的生成或分泌有关。OBJECTIVE To explore the therapeutic effects of Qingdu Shenjin Granules(QSG)on adjuvant arthritis(AA)rats.METHODS Male Sprague-Dawley(SD)rats were utilized for AA rat modeling with Freund’s complete adjuvant.Then the animals were randomized into five groups of model,Wantong Jingu Tablet(0.15 g·kg^(–1))and QSG low/medium/high-dose(3.9,7.8,15.6 g·kg^(–1))(n=10 each).Another 10 normal SD rats were designated as normal control group.Each group a daily gavage for 30 consecutive days.Pain threshold,foot swelling degree and arthritis score were measured regularly.After dosing,serum immunoglobulin G(IgG),immunoglobulin M(IgM),tumor necrosis factor alpha(TNF-α)and interleukin-1 beta(IL-1β)were detected by enzyme-linked immunosorbent assay(ELISA).Joint pathological injury was examined after hematoxylin-eosin stain of joint sections.And histopathological changes of ankle joint were scored.RESULTS Compared with normal control group,pain threshold of AA rats declined markedly.Foot swelling degree,arthritis index and serum levels of inflammatory cytokines spiked markedly.Synovial layer,cartilage and bone layer of ankle joint became severely injured and there was a marked rise of ankle joint pathological score(P<0.05,P<0.01).As compared with model group,QSG could significantly elevate the pain threshold of AA rats,improve foot swelling,reduce arthritis index,lower serum levels of inflammatory cytokines,lessen the injury of synovial layer,cartilage and bone layer of ankle joint and lower ankle joint pathological tissue score(P<0.05,P<0.01).CONCLUSION QSG has therapeutic effect on AA rats.And its mechanism may be correlated with lowering the production or secretion of inflammatory cytokines.
