摘  要:目的:探讨姜黄素对肠易激综合征(IBS)大鼠直肠黏膜嗜铬细胞和血清5-羟色胺(5-HT)水平的影响。方法:将30只SD大鼠随机分为对照组(6只)和造模组(24只),对照组不造模,造模组采用多因素慢急性联合应激形态构建动物模型。造模成功后将造模组大鼠随机分为模型组,姜黄素低、中、高剂量组,每组6只。造模结束后,姜黄素低、中、高剂量组分别将10mg/kg、20mg/kg、40mg/kg的姜黄素溶于2mL生理盐水中,制备相应剂量的药液进行灌胃。对照组和模型组大鼠给予2mL常温生理盐水灌胃。各组灌胃21d。观察对照组和造模组大鼠体重、粪便粒数、粪便含水量情况,统计各组直肠黏膜嗜铬细胞数量、血清5-HT水平。结果:对照组和模型组大鼠造模后体重比较,差异无统计学意义(P>0.05)。造模组12 h粪便粒数、粪便含水量较造模前和对照组明显增加,差异有统计学意义(P<0.05)。模型组与姜黄素低、中、高剂量组疼痛阈值较对照组明显降低,血清5-HT水平和直肠黏膜嗜铬细胞数量明显高于对照组,差异有统计学意义(P<0.05)。姜黄素低剂量组与模型组疼痛阈值、直肠黏膜嗜铬细胞数量比较,差异无统计学意义(P>0.05);姜黄素低剂量组血清5-HT水平显著低于模型组,差异有统计学意义(P<0.05)。姜黄素中、高剂量组疼痛阈值显著高于模型组与姜黄素低剂量组,血清5-HT水平、直肠黏膜嗜铬细胞数量显著低于模型组与姜黄素低剂量组,差异有统计学意义(P<0.05)。姜黄素中剂量组疼痛阈值与姜黄素高剂量组比较,差异无统计学意义(P>0.05);姜黄素高剂量组血清5-HT水平、直肠黏膜嗜铬细胞数量显著低于姜黄素低剂量组,差异有统计学意义(P<0.05)。结论:姜黄素可以显著降低IBS大鼠嗜铬细胞数量和血清5-HT水平,改善直肠敏感性,促进大鼠肠道功能恢复。Objective:To investigate the effects of curcumin on rectal mucosal chromaffin cells and serum 5-hydroxytryptamine(5-HT)levels in rats with irritable bowel syndrome(IBS).Methods:Thirty SD rats were randomly divided into a control group(six rats)and a modeling group(twenty-four rats),the control group was not modeled,and the modeling group was constructed using a multifactorial slow-acute combined stress pattern to build an animal model.After successful modeling,the rats in the modeling group were randomly divided into model group,curcumin low-,medium-and high-dose groups,6 rats in each group.At the end of the modeling,10 mg/kg,20 mg/kg and 40 mg/kg of curcumin were dissolved in 2 mL of saline in the curcumin low-,medium-and high-dose groups,respectively,and the corresponding dosage of curcumin was prepared by gavage.In the control and model groups,rats were gavaged with 2 mL of saline at room temperature.Each group was gavaged for 21 d.The body weights,fecal pellet counts and fecal water contents of the rats in the control and model groups were observed,and the numbers of chromophores in the rectal mucosa and the serum 5-HT levels of the rats in each group were counted.Results:There was no statistically significant difference in the body weight of rats in the control group and the model group after modeling(P>0.05);the number of fecal pellets and fecal water content in the model group at 12 h increased significantly compared with those in the pre-modeling and control groups,and the difference was statistically significant(P<0.05);the pain threshold of the modeling group and curcumin low,medium and high dose group was significantly lower than that of the control group,and the serum 5-HT level and the number of rectal mucosal chromophores were significantly higher than that of the control group,and the difference was statistically significant(P<0.05);the difference in the pain threshold and the number of rectal mucosal chromophores of the low dose group of curcumin compared with that of the models was not statistica
