食欲素受体激动剂研究进展

摘　　要：食欲素是下丘脑分泌的兴奋性神经肽,具有广泛的生理作用,包括调节摄食和能量平衡、调节睡眠-觉醒状态、调节成瘾性、调节焦虑及抑郁等。目前关于食欲素受体拮抗剂在治疗失眠方面的研究报道较多,并制作成药物上市。相比于食欲素受体拮抗剂的研究来说,食欲素受体激动剂的研究相对较少,但其发展前景不容忽视,本文综述了从食欲素发现以来有关食欲素受体激动剂的研究,发现其在治疗发作性睡病、逆转药物性呼吸抑制、促进肿瘤细胞凋亡等方面的潜在作用。通过综述近年来的研究成果以及对目前存在问题的讨论以期为开发新药提供理论依据和研究思路。Orexin is an excitatory neuropeptide secreted by the hypothalamus,with many physiological effects,including regulating food intake and energy balance,sleep-wake state,addiction,anxiety and depression,etc.At present,only the complete study of orexin receptor antagonists for insomnia has been reported and made into drugs.Compared with orexin receptor antagonists,research on orexin receptor agonists is still very few,despite its prospect in development.This article reviewed the research on orexin receptor agonists,their potential roles in the treatment of narcoleptic disease,reversal of drug-induced respiratory depression,and promotion of cancer cell apoptosis.This article also reviewed the research in recent years and discussed the existing problems to provide theoretical basis and ideas for the development of new drugs.
