摘 要:以韭菜迟眼蕈蚊的转录组数据为基础,通过RT-PCR克隆并获得韭菜迟眼蕈蚊的两个气味受体基因BodoOR71和BodoOR72的cDNA全长序列,对其序列及结构特征进行生物信息学分析,比较二者在韭菜迟眼蕈蚊不同发育时期及成虫不同组织中的表达量差异。序列分析发现,BodoOR71的开放阅读框(open reading frame,ORF)为759 bp,编码253个氨基酸,具有4个跨膜结构域;BodoOR72的ORF为954 bp,编码318个氨基酸,具有6个跨膜结构域。系统进化树结果表明,BodoOR71和BodoOR72与其他双翅目昆虫亲源关系较近,能够聚在一支。时空表达模式结果表明,BodoOR71在除雄成虫之外的不同虫态均有表达,且在雌成虫中的表达量最高,在雌成虫触角中的表达量显著高于其他组织,在雄成虫各组织中均不表达;BodoOR72在不同虫态均有表达,且在雄成虫中的表达量最高,在雄成虫触角中的表达量显著高于其他组织,在雌成虫各组织中微量表达。推测BodoOR71和BodoOR72可能参与韭菜迟眼蕈蚊嗅觉识别过程。Based on the transcriptome data of Bradysia odoriphaga,the full-length cDNA sequences of BodoOR71 and BodoOR72 were cloned by RT-PCR,sequence as well as structure analysis were done by bioinformatic tools,and comparison between expression levels of these genes in different developmental stages and adult tissues were done.The sequence analysis showed that BodoOR71 had an open reading frame(ORF)of 759 bp,encoding a protein of 253 amino acids with four transmembrane domains,and BodoOR72 had an ORF of 954 bp,encoding a protein of 318 amino acids with six transmembrane domains.The phylogenetic analysis indicated that BodoOR71 and BodoOR72 were closely related to other Diptera insects,which can be clustered into one clade.The temporal and spatial expression pattern showed that BodoOR71 was expressed at different developmental stages except males and highly expressed in females.The expression level of BodoOR71 was significantly higher in female antennae than other tissues and was barely detected in males.BodoOR72 was expressed at different developmental stages and highly expressed in males.The expression level of BodoOR72 stayed low in all tissues of female while turned significantly higher in male antennae than other tissues.To conclude,BodoOR71 and BodoOR72 may involve in the olfactory recognition of B.odoriphaga.
