添加牛樟和阴香基质对高黎贡牛樟芝萜类化合物生物合成途径的影响

摘  要:为了研究添加牛樟和阴香嫩枝叶粉末对高黎贡牛樟芝萜类化合物的影响,使用Illumina高通量测序平台对添加牛樟、阴香嫩枝叶和薏仁(对照组)粉末培养液体培养后的菌丝体进行转录组测序,并分析萜类化合物合成途径关键基因表达情况。结果显示:(1)在添加牛樟枝叶粉末(BSNZZ-NF)和阴香枝叶粉末(BSNZZ-YF)下,共有446个差异表达基因,261个上调、185个下调;对差异表达基因的GO富集分析发现,与氧化还原过程及各种氧化还原酶相关的基因被显著富集;KEGG pathway富集分析发现,DEGs显著富集在酪氨酸、乙醛酸和二羧酸、丙酸三个代谢通路中。(2)在高黎贡牛樟芝转录组数据中,共筛选出43条参与萜类化合物合成的甲羟戊酸(MVA)途径的转录序列,添加牛樟基质后,HBD9、HMG-CoA、GGT3和OCS表达量明显上调,HBD12、TPS5和SE基因表达量下调;添加阴香基质后,AACT1、HBD5、HBD8、HBD12、FPPS1表达量明显上调,HMGS、HMG-CoA、IDI表达量下调。(3)有bHLH、bZIP、MYB等14个转录因子与高黎贡牛樟芝萜类化合物生物合成相关,14个转录因子在添加牛樟和阴香粉末后均具有不同程度的表达。本研究为进一步揭示参与高黎贡牛樟芝萜类生物合成相关转录因子反应机制提供了参考。Taiwanofungus camphoratus is a rare edible and medicinal fungus.Its main components terpenoids have anticancer effects.In order to study the effects of adding Cinnamomum kaneirae and C.burmanni leaves matrix on the terpenoids of T.gaoligongensis,this study used Illumina high-flux sequencing platform to perform transcriptome sequencing on the mycelium of C.kaneirae,C.burmanni leaves and Coicis Semen(control group)matrix cultured in liquid medium,and the expression of key genes in the pathway of terpenoid synthesis were analyzed.The results showed that there were 446 differentially expressed genes,261 up-regulated and 185 down-regulated,when BSNZZ-NF and BSNZZ-YF were added.The GO enrichment analysis of DEGs showed that the genes related to oxidation-reduction process and various oxidoreductase activity were significantly enriched.KEGG pathway enrichment analysis showed that DEGs was significantly enriched in three metabolic pathways:Tyrosine metabolism,Propanoate metabolism,Glyoxylate and dicarboxylate metabolism.In the transcriptome data of T.gaoligongensis,a total of 43 mevalonate pathway(MVA pathway)involved in the synthesis of terpenoids were screened.After adding C.kaneirae,the expressions of HBD9,HMG-CoA,GGT3 and OCS were significantly up-regulated,while the expressions of HBD12,TPS5 and SE genes were down-regulated.The expression levels of AACT1,HBD5,HBD8,HBD12 and FPPS1 were significantly up-regulated,while the expression levels of HMGS,HMG-CoA and IDI were down-regulated after the addition of C.burmanni.There were 14 transcription factors,including bHLH,bZIP and MYB,which were related to the biosynthesis of terpenoid compounds of T.gaoligongensis.The 14 transcription factors were expressed in different degrees after adding C.kaneirae and C.burmanni matrix.This study provides a reference for further elucidating the mechanism of transcription factors involved in the terpenoid biosynthesis of T.gaoligongensis.
