AgBr纳米微粒和阳离子共聚物修饰的壳聚糖基抗菌复合物的制备及抗菌活性研究

摘  要:双重抗菌机制的多孔状抗菌复合材料相较于仅具有单一抗菌机制的一维材料而言,具有更大的比表面积和高效稳定的抗菌活性,这是其重要的优势。首先以丙烯酸丁酯(BA)、甲基丙烯酸缩水甘油酯(GMA)和对苯乙烯磺酸钠(SPS)作为聚合单体,合成了两亲性的聚丙烯基(PBGS)共聚物。然后,将得到的PBGS共聚物与壳聚糖(CS)通过共价键交联,继而合成了壳聚糖/三元共聚物(PBGS-C)复合物。合成的PBGS-C复合物再与十六烷基三甲基溴化铵(CTAB)发生离子交换反应,制备得到了具有季铵阳离子基团的壳聚糖/PBGS共聚物(PBGS-C/N)抗菌复合物。最后,采用银离子对PBGS-C/N抗菌复合物继续功能化的方法,合成了具有接触/释放双重抗菌机制的AgBr纳米微粒和阳离子共聚物修饰的(AgBr@PBGS-C/N)抗菌复合物。AgBr@PBGS-C/N抗菌复合物的组成和形貌通过红外光谱(FT-IR)、X光电子能谱(XPS)、扫描电镜(SEM)和能谱(EDS)等测试进行确认。此外,它的抗菌活性通过大肠杆菌和金黄色葡萄球菌进行了评估。结果表明:AgBr@PBGS-C/N抗菌复合物具有三维多孔结构和良好的抗菌活性,有望用于环保、医疗、食品加工等领域。Porous antibacterial composites with dual antibacterial mechanisms have larger surface area and high-efficient and stable antibacterial activity compared to one-dimensional materials with a single antibacterial mechanism,which is their important advantages.In this study,an amphiphilic polyacrylate-based(PBGS)copolymer was first synthesized using butyl acrylate(BA),glycidyl methacrylate(GMA)and sodium p-styrene sulfonate(SPS)as polymerization monomers.Afterwards,a chitosan/terpolymer(PBGS-C)composite was synthesized by crosslinking the obtained PBGS copolymer with chitosan(CS)through covalent bonds.The synthesized PBGS-C composite then underwent an ion exchange reaction with cetyltrimethyl ammonium bromide(CTAB),and the quaternary ammonium groups-functionalized(PBGS-C/N)antibacterial composite was further prepared.Finally,AgBr nanoparticles and cationic copolymer modified chitosan-based(AgBr@PBGS-C/N)antibacterial composite with contact/release dual antibacterial mechanisms was successfully synthesized by further functionalization of PBGS-C/N composite using silver ions.The composition and morphology of the AgBr@PBGS-C/N antibacterial composite were confirmed by FT-IR,XPS,SEM and EDS tests.In addition,its antibacterial activity was evaluated against Escherichia coli(E.coli)and Staphylococcus aureus(S.aureus).The results showed that AgBr@PBGS-C/N antibacterial composite had three-dimensional porous structure and favorable antibacterial activity,which was expected to be used in environmental protection,medical treatment,food processing and other fields.
