摘 要:TCP转录因子家族在叶片发育中发挥重要作用,鸭茅是世界著名的多年生禾本科牧草,叶片是其主要的收获部位,且营养价值丰富。本研究鉴定了鸭茅(Dactylis glomerata)TCP基因家族成员,并对所有成员进行了理化性质、基因结构、染色体定位、进化关系、保守基序和结构域、启动子顺式作用元件以及表达量等分析。发现鸭茅TCP基因家族共有22个成员被划分成3个亚家族;它们都含有TCP蛋白典型结构域;成员间蛋白的亲水性较好,等电点和相对分子质量有差异;α螺旋和无规则卷曲是鸭茅TCP蛋白主要的二级结构,亚细胞定位预测主要定位于细胞核;22个成员的启动子顺式作用元件可分为4类,分别是光响应元件、植物激素响应元件、抗病创伤胁迫响应元件以及其他元件;此外,DgTCP8和DgTCP20在叶片中的表达量较高且可响应生长素和赤霉素的表达。本文为深入探究鸭茅TCP基因在叶片发育中的功能提供依据,为叶片优良鸭茅品种的选育奠定基础。The TCP transcription factor family genes play an important role in leaf development,and orchardgrass is a world-renowned perennial Poaceae grass.Its leaves are the main harvesting parts and have rich nutritional value.In this study,we identified the members of the TCP gene family in orchardgrass and analyzed their physicochemical properties,gene structure,chromosome localization,evolutionary relationships,conserved motifs and domains,cis-acting promoter elements,and expression levels.It was found that 22 members of the TCP gene family in orchardgrass were divided into 3 groups.They all contain typical domains of TCP proteins.The hydrophilicity of proteins among members is good,and there were differences in isoelectric point and relative molecular weight.α-helix and random coil were the main secondary structures of orchardgrass TCP protein,and subcellular localization prediction was mainly located in the nucleus.The cis acting elements of the 22 member promoters can be divided into 4 categories,namely light responsive elements,plant hormone responsive elements,anti traumatic stress responsive elements,and other elements.In addition,DgTCP8 and DgTCP20 had higher expression levels in leaves and responded to treatment with auxin and gibberellin.The article provides a basis for in-depth exploration of the function of the TCP gene in leaf development of orchardgrass,and lays the foundation for the breeding of excellent orchardgrass varieties.
