摘  要:目的以他克莫司为模型药,考察不同牛血清替代人血清作为空白基质在HPLC-MS/MS药物浓度测定中的可行性研究。方法在相同的处理方法下,以人血清和不同牛血清作为空白基质制备标准曲线,对比101份他克莫司血清样品在不同标准曲线下的浓度是否存在差异。结果三种牛血清(小牛血清、新生牛血清、胎牛血清)与人血清的相对偏差均大于TEa(12.5%),均不能替代人血清作为空白基质。结论以他克莫司为模型药时,在HPLC-MS/MS法测定药物浓度中,三种牛血清均不能替代人血清作为空白基质。Objective Using tacrolimus as the model drug,the feasibility analysis of different bovine serum replacing human serum as blank substrate was investigated in the determination of drug concentration by HPLC-MS/MS.Methods Under the same treatment method,human serum and different bovine serum were used as blank matrix to prepare standard curves,and the concentration of 101 tacrolimus serum samples was compared under different standard curves.Results The relative deviation between the three types of bovine serum(calf serum,newborn bovine serum,fetal bovine serum)and human serum was greater than TEa(12.5%),and they could not replace human serum as blank substrate.Conclusion When tacrolimus was used as the model drug,none of the three bovine serum could replace human serum as the blank substrate in the determination of drug concentration by HPLC-MS/MS.
