摘 要:目的观察蒙药胡日查-6和古日古木-13对变应性鼻炎(allergic rhinitis,AR)小鼠的鼻黏膜病变及血清中IgE、IL-4和IFN-γ含量的影响。方法将50只健康BALB/c小鼠,雌雄各半,分为空白对照组、AR模型组、西替利嗪组、胡日查-6组及古日古木-13组。建立AR小鼠模型,对每个实验组的小鼠行为学评分,随机抽取空白组、造模组小鼠各1只进行被动皮肤过敏原实验(passive skin allergen experiment,PCA)。在给药治疗后,观察小鼠鼻黏膜病理组织学变化,测定血清中IgE、IL-4和IFN-γ的含量。结果(1)造模成功的AR模型小鼠出现剧烈挠鼻、打喷嚏等症状,且评分≥5分;经治疗后,各给药组鼻部症状有所减轻,症状评分有所下降。(2)抽取的模型组小鼠PCA实验可见阳性蓝斑,空白组未见蓝斑形成,佐证模型组小鼠造模成功。(3)AR模型组鼻黏膜组织学观察可见较多炎性细胞浸润,间质增宽,小血管扩张、增多。经治疗后,各给药组小鼠症状有所缓解,鼻黏膜组织中炎症细胞减少,其中以胡日查-6组最为明显。(4)模型组血清IgE、IL-4的含量相较于空白对照组明显升高(F=95.64,F=238.71,P<0.01),IFN-γ的含量较低(F=54.25,P<0.05)。经治疗后各给药组小鼠鼻部症状均有所减轻,IgE和IL-4的含量较模型组明显下降(F=28.60,F=25.22,P<0.01),IFN-γ的含量升高(F=17.04,P<0.05)。蒙药各组和西替利嗪组比较,IgE差异无统计学意义(F=0.098,P>0.05),IL-4差异无统计学意义(F=0.316,P>0.05),IFN-γ有统计学差异(F=5.177,P<0.05),各组性别上无明显差异(P>0.05)。结论蒙药胡日查-6和古日古木-13能够有效改善变应性鼻炎模型小鼠挠鼻、喷嚏、流涕以及精神不济等典型症状,促进鼻黏膜修复,减轻AR小鼠病理损伤;通过调节免疫相关细胞因子IgE、IL-4和IFN-γ,从而有效治疗变应性鼻炎,为探索蒙药治疗AR的相关机理奠定理论基础,为未来治疗AR靶点的筛选及靶向治疗提供研究依据。Objective To observe the effects of Mongolian Huricha-6 and Gurigumu-13 on nasal mucosal lesions and IgE,IL-4 and IFN-γcontent in allergic rhinitis(AR)mice.Methods 50 healthy BALB/c mice were divided into blank control group,AR model group,cetirizine group,Huricha-6 group and Gurigumu-13 group.AR mouse model was established,the behavioral score of each experimental group was scored,and one blank group and one mouse were randomly selected for passive skin allergen experiment(PCA).After treatment,administration,the pathological histological changes in the nasal mucosa were observed and the amounts of IgE,IL-4 and IFN-γin serum were determined.Results(1)The successful AR model mice developed symptoms such as severe nose scratching and sneezing,and scored 5 points;after treatment,the nasal symptoms decreased and the symptom score decreased.(2)The PCA of the mice in the selected model group was positive,and no blue formation was found in the blank group,assuring the successful modeling of the mice in the model group.(3)Histological observation of the nasal mucosa in the AR model group showed more inflammatory cell infiltration,more interstitial widening,and dilated and more small blood vessels.After treatment,the symptoms of mice in each treatment group were relieved and inflammatory cells were decreased in the nasal mucosa,among which the Huricha-6 group was the most significant.(4)The serum contents of IgE and IL-4 in the model group were significantly higher compared with the blank control group,and the content of IFN-γwas relatively low(P<0.05).After treatment,the nasal symptoms and the content of IgE and IL-4 decreased significantly compared with the model group,and the content of IFN-γincreased(P<0.05).Between each group and cetirizine,there was no significant difference in IgE(F=0.098,P>0.05),IL-4(F=0.316,P>0.05),IFN-γwas statistically different(F=5.177,P<0.05),and no gender difference in each group(P>0.05).Conclusion Mongolian Huricha-6 and Gurigumu-13 can effectively improve the typical symptoms such as
