摘 要:目的:比较补阳还五汤与黄芪甲苷治疗大鼠缺血性脑卒中的疗效。方法:将50只SD大鼠随机分组,分为假手术组(Sham组)、模型组(MCAO组)、补阳还五汤组(BYHWD组)、黄芪甲苷组(AS-IV组)与血塞通组(XueSaiTong组),每组各10只。建立大鼠大脑中动脉闭塞(middle cerebral arterial occlusion,MCAO)模型,利用Zea Longa法评估神经功能缺损程度,TTC染色检测脑梗死体积,HE染色观察神经细胞病理形态学改变,尼氏染色观察尼氏体变化,免疫组化染色、免疫荧光染色观察和比较神经元特异性核蛋白NeuN表达。结果:与MCAO组相比,BYHWD组、AS-IV组和XueSaiTong组大鼠神经功能缺损症状均减轻(P<0.05),脑梗死体积缩小(P<0.05),神经细胞形态规则,尼氏体数量增多(P<0.05),NeuN蛋白表达升高(P<0.05);与AS-IV组相比,BYHWD组神经功能缺损症状有所减轻,但差异无统计学意义(P>0.05),其余检测结果以BYHWD组疗效较好(P<0.05)。结论:补阳还五汤、黄芪甲苷二者均可起到减轻MCAO大鼠神经功能缺损的效果,发挥神经保护作用;其中补阳还五汤治疗效果更好。Objective:To compare the therapeutic effects of Buyang Huanwu Decoction and Astragaloside IV in the treatment of ischemic stroke in rats.Methods:Fifty SD rats were randomly divided into five groups:sham operation group(Sham group),middle cerebral artery occlusion model group(MCAO group),Buyang Huanwu Decoction group(BYHWD group),Astragaloside IV group(AS-IV group),and Xue Sai Tong group,with 10 rats in each group.The middle cerebral artery occlusion(MCAO)model was established in rats.The Zea Longa method was used to assess the degree of neurological cleficit,TTC staining to detect the volume of cerebral infarction;HE staining to observe pathological morphological changes in nerve cells;Nissl staining to observe changes in Nissl bodies;Immunohistochemical staining and immunofluorescence staining to observe and compare the expression of neuron-specific nuclear protein NeuN.Results:Compared with the MCAO group,both the BYHWD group and the AS-IV group showed improvements in neurological deficit symptoms(P<0.05),reduced cerebral infarction volume(P<0.05),regular neuronal cell morphology,increased Nissl body counts(P<0.05),and elevated NeuN protein expression(P<0.05).When compared with the AS-IV group,the BYHWD group displayed a slight reduction in neurological deficit symptoms,but the difference was not statistically significant(P>0.05).However,the BYHWD group showed more significant therapeutic effects in other detected parameters(P<0.05).Conclusion:Both BYHWD and Astragaloside IV can alleviate the neurological deficit in MCAO rats,exerting neuroprotective effects.Among them,BYHWD exhibits the most significant therapeutic effect.
