摘 要:采用3-脱氢莽草酸乙酯为原料通过缩合芳构化反应制备得到一种粘度响应近红外荧探针L,采用核磁、红外、高分辨质谱等表征方法对探针结构进行确证。结果表明:探针L在高粘度溶剂中表现出显著的荧光增强现象,而在低粘度溶剂中荧光极其微弱,证实该探针具有粘度响应特征;此外,探针L还具有良好的抗干扰性、pH稳定性及光稳定性;利用探针L的粘度-荧光响应性能,对脂多糖(LPS)及制霉菌素(Nystatin)导致的HeLa细胞粘度变化进行了荧光成像。Probe L was synthesized using ethyl 3-dehydroshikimate as the raw material through an aromatic condensation reaction.The structure of probe L was confirmed using magnetic resonance imaging,infrared spectroscopy and high-resolution mass spectrometry.Probe L exhibited significant fluorescence enhancement in high-viscosity solvents such as glycerol and extremely weak fluorescence in low-viscosity solvents,confirming its notable viscosity response feature.Moreover,probe L demonstrated good anti-interference capability,high pH stability and good photo-stability.Leveraging the viscosity-fluorescence response of probe L,fluorescence imaging of viscosity changes in HeLa cells induced by LPS or Nystatin was conducted.
