苯甲腈降解菌Rhodococcus sp.BN02的筛选和鉴定

摘  要:本研究从武汉市东西湖区金银湖附近水体污泥中提取以苯甲腈为唯一碳氮源,筛选出一株可高效降解腈类化合物的菌株。经生理生化测定、16S rRNA基因序列测定、比对和系统发育树构建,确认该菌为红球菌属(Rhodococcus),并命名为Rhodococcus sp.BN02。利用单因素实验和响应面实验对Rhodococcus sp.BN02以苯甲腈为唯一碳氮源的生长特性进行研究,发现其在28℃、pH 7和800 mg/L苯甲腈的条件下生长最适宜,最大OD 600值可达0.471,并在2 d内可将苯甲腈完全降解。此外,本研究还考察了BN02对不同腈类化合物的降解能力,它在48 h内可完全降解500 mg/L苯甲腈,在4 d内可完全降解500 mg/L的乙腈、丙烯腈和琥珀腈,对500 mg/L的2-氰基吡啶培养7 d后降解率可达83.84%。本研究为消除环境中苯甲腈及其中间代谢有毒污染物以及进一步探究苯甲腈代谢机制提供理论依据。In this study,a strain capable of efficiently degrading nitrile compounds was isolated from the sludge near Jinyin Lake in Dongxihu District,Wuhan City,China,using benzonitrile as the sole carbon and nitrogen source.The strain was identified as belonging to the Rhodococcus and was named Rhodococcus sp.BN02 through physiological and biochemical measurements,16S rRNA gene sequencing,sequence alignment and phylogenetic tree construction.Single-factor and response surface analysis were conducted to study the growth characteristics of Rhodococcus sp.BN02 when benzonitrile was the sole carbon and nitrogen source.It was found that the strain grew optimally at 28℃,pH 7,and with 800 mg/L of benzonitrile,achieving a maximum OD 600 value of 0.471 and complete degrading of benzonitrile with in 2 d.In addition,the ability of BN02 to degrade different nitrile compounds was investigated.It was observed that BN02 completely degraded 500 mg/L benzonitrile within 48 h,and 500 mg/L acetonitrile,acrylonitrile,and succinonitrile within 4 d.Moreover,for 500 mg/L of the 2-cyanopyridine,the degradation rate after 7 days of cultivation reached 83.84%.This study provided a theoretical basis for the elimination of benzonitrile and its intermediate metabolic pollutants in the environment,as well as further exploration of the metabolic mechanism of benzonitrile.
