摘  要:【目的】探究β_(3)-肾上腺素受体(β_(3)-AR, ADRB3)在犊牦牛寒冷适应中的调控作用。【方法】选取不同日龄(1、7和30日龄)的健康雄性犊牦牛,采用免疫组织化学染色(IHC)、实时荧光定量PCR(qRT-PCR)以及Western blot(WB)对犊牦牛棕色/白色脂肪组织中ADRB3 mRNA及β_(3)-AR蛋白的表达、分布及激素敏感性脂肪酶(HSL)基因mRNA表达进行分析。【结果】IHC结果显示,β_(3)-AR在犊牦牛肾周棕色和白色脂肪细胞中均有分布,且从1~30日龄,β_(3)-AR阳性表达强度显著增加(P<0.05)。qRT-PCR结果显示,从1~30日龄,ADRB3和HSL mRNA表达水平显著增加(P<0.05)。WB结果显示,从1~30日龄,β_(3)-AR蛋白表达水平显著增加(P<0.05)。【结论】ADRB3 mRNA和β_(3)-AR蛋白在犊牦牛肾周棕色和白色脂肪组织中均有表达,且其表达量在白色脂肪组织中高于棕色脂肪组织,说明β_(3)-AR在犊牦牛肾周脂肪组织产热和脂解中均发挥重要的调节作用。【Objective】The aim of this study was to explore the regulatory role ofβ_(3)-adrenergic receptor(β_(3)-AR,ADRB3)in cold adaptation of calf yaks.【Method】Healthy male calf yaks of different ages(1,7 and 30 days)were selected.The expression and distribution of ADRB3 mRNA,β_(3)-AR protein and the hormone-sensitive triglyceride lipase(HSL)mRNA in the perirenal brown and white adipose tissue of calf yaks were detected by immunohistochemical staining(IHC),Real-time fluorescence quantitative PCR(qRT-PCR),and Western blot(WB).【Result】The IHC results showed that theβ_(3)-AR was dis⁃tributed both in perirenal brown and white adipocytes of yak calves,and the positive expression intensity ofβ_(3)-AR was significantly increased from day 1 to 30(P<0.05).The results of qRT-PCR showed that the mRNA expression level of ADRB3 and HSL were significantly increased from day 1 to 30(P<0.05).The re⁃sults of WB showed that the protein expression level ofβ_(3)-AR were significantly increased from day 1 to 30(P<0.05).【Conclusion】ADRB3 mRNA andβ_(3)-AR protein were expressed in both perirenal brown and white adipocytes of yak calves,and its expression level in white adipose tissue were higher than that in brown adipose tissue,indicating thatβ_(3)-AR plays an important regulatory role in nonshivering thermogen⁃esis(NST)and lipolysis in perirenal adipose tissue of yak calves.
