摘 要:目的:基于谱效关系研究并经实验验证,阐明独一味抗炎药效物质基础。方法:对12批不同来源的独一味进行HPLC分析,建立指纹图谱。基于小鼠耳肿胀模型,测定各批次独一味水提物的耳肿胀率及其抑制率、血清中TNF-α、IL-6的含量。运用灰色关联度法及文献对比,分析独一味水提物中与抗炎药效关联较强的成分,验证其在独一味水提物抗炎药效中的贡献度。结果:建立了独一味水提物HPLC指纹图谱,共标定了16个共有峰,通过对照品指认,辨识了山栀子苷甲酯、8-O-乙酰山栀子苷甲酯、连翘酯苷B和木犀草苷等7个特征峰成分,除胡麻属苷、芦丁含量较低外,其余5种成分均有较高含量。二甲苯致耳肿胀试验结果表明,12批独一味水提物均有抑制耳肿胀作用。通过灰色关联度法和文献对比研究分析,推测山栀子苷甲酯、8-O-乙酰山栀子苷甲酯、连翘酯苷B、木犀草苷与独一味水提物抗炎作用密切相关,对此4种成分进行含量测定,含量测定结果依次为(15.319±4.228)、(26.156±7.174)、(14.376±5.817)、(12.756±2.570)mg/g。以对照品模拟活性成分配比构建药效对比验证实验确认了这一推测。结论:山栀子苷甲酯、8-O-乙酰山栀子苷甲酯、连翘酯苷B、木犀草苷可能为独一味水提物抗炎药效的关键成分群。Objective:To elucidate the anti-inflammatory material basis of Lamiophlomis rotata(Benth.)Kudo(L.rotata)based on the spectrum-effect relationship and experimental validation.Methods:The fingerprint of 12 batches of L.rotata from different origins was established based on HPLC analysis.Based on mouse ear swelling model,the ear swelling rate and inhibition rate in each batch of L.rotata was measured,and the serum contents of TNF-αand IL-6 were detected.Based on gray correlation analysis and literature comparison,the key ingredients of L.rotata against inflammation were investigated and their contribution to the anti-inflammatory effect of the L.rotata water extract was verified.Results:HPLC fingerprint of L.rotata water extract was established with 16 common peaks identified.Seven characteristic peaks were identified by reference substances,such as shanzhiside methylester,8-O-acetylshanzhiside methyl ester,forsythiaside B,luteolin-7-O-glucoside,and so on.Except for sesamoside and rutin,high levels were observed in the other 5 peaks.The results of ear swelling induced by xylene showed all 12 batches of L.rotata water extract had anti-inflammatory effects.The results of gray correlation analysis and literature comparison suggested that shanzhiside methylester,8-O-acetylshanzhiside methyl ester,forsythiaside B,luteolin-7-O-glucoside were closely related with the anti-inflammatory effect of L.rotata water extract.The contents of these four components were(15.319±4.228),(26.156±7.174),(14.376±5.817),(12.756±2.570)mg/g,respectively.Further pharmacological comparison validation confirmed the speculation.Conclusion:Shanzhiside methylester,8-O-acetylshanzhiside methyl ester,forsythiaside B,luteolin-7-O-glucoside might be the key ingredients of L.rotata water extract against inflammation.
