摘 要:目的研究复方MYZ-20对耐药铜绿假单胞菌的抑制作用及机制。方法采用倍比稀释法确定MYZ-20的最低抑菌浓度和最小杀菌浓度;通过检测相对电导率、胞外碱性磷酸酶活性确定MYZ-20的杀菌功效;测定绿脓素、绿色荧光素、生物膜形成,探究MYZ-20对耐药铜绿假单胞菌毒力因子的抑制作用;建立小鼠烫伤感染模型,观察MYZ-20抗感染与创口修复的效果。结果MYZ-20通过破坏膜结构杀伤铜绿假单胞菌,显著降低细菌毒力因子水平,促进小鼠伤口修复及降低炎症水平。结论MYZ-20对耐药铜绿假单胞菌具强杀伤作用,并具有抗炎及伤口修复作用。OBJECTIVE To study the inhibitory effect and mechanisms of compound MYZ-20 on drug-resistant Pseudomonas aeruginosa.METHODS The minimum inhibition concentration and the minimum bactericidal concentration of MYZ-20 were determined by the double dilution method.The bactericidal efficacy of MYZ-20 was confirmed by measuring the relative conductivity and extracellular alkaline phosphatase activity,and the inhibitory effect of MYZ-20 on the virulence factors of drug-resistant P.aeruginosa was explored by testing the pyocyanin,green fluorescein,and the formation of biofilm.Additionally,a scalded infection mouse model was established for the determination of the anti-infection and wound healing effects ofMYZ-20.RESULTSS MYZ-20 killed P.aeruginosaby destroying the membrane structure,significantly reducing the level of bacterial virulence factors,promoting wound repair and reducing the level of inflammation in mice.CONCLUSION MYZ-20 has a strong bactericidal effect on drug-resistant P.aeruginosa,and has anti-inflammatory and wound repair effects.
