摘 要:为探究基于晚期糖基化终末产物受体(RAGE)和血管细胞粘附分子1(VCAM-1)表达变化的抗静脉血栓方对深静脉血栓(DVT)大鼠抗血栓的影响,将30只SD大鼠随机分为空白组、模型组、假手术组、阳性药组和中药组,采用下腔静脉狭窄法建立大鼠DVT模型。中药组给予抗静脉血栓方汤剂4.48 g/100 g;阳性药组给予复方丹参片0.023 g/100 g;其余组灌胃等体积生理盐水,1次/d。灌胃7 d后,取腹主动脉血1~2 mL,取下腔静脉标本。ELISA检测大鼠血清中RAGE和VCAM-1的表达水平,HE染色观察血管形态改变,免疫组化、WB和PCR检测各组大鼠RAGE和VCAM-1的表达变化。结果显示:模型组大鼠内膜损伤严重,有血栓形成,血管壁有炎性细胞浸润;模型组大鼠血清RAGE和VCAM-1的含量均显著高于中药组和阳性药组,而中药组和阳性药组大鼠血管组织中RAGE和VCAM-1的表达均降低;与假手术组相比,模型组大鼠中的RAGE、VCAM-1的蛋白和mRNA表达均显著增加,药物治疗后,中药组与阳性药组RAGE\,VCAM-1的蛋白和mRNA表达均显著降低。研究表明排除手术创伤影响,抗静脉血栓方可能是通过减少RAGE、VCAM-1的表达从而减少DVT模型大鼠静脉血栓的形成。The purpose of this paper is to investigate the effect of anti-venous thrombosis decoction based on RAGE and VCAM-1 expression on antithrombotic effect in rats with deep vein thrombosis(DVT).30 SD rats were randomly divided into blank group,model group,sham operation group,positive drug group and Chinese traditional medicine group.DVT model was established by inferior vena cava stenosis method.The Chinese traditional medicine group was given anti-venous thromosis decoction 4.48 g/100 g;the positive drug group was given Compound Danshen Tablet 0.023 g/100 g;the other groups were given equal volume of normal saline once a day.After 7 days of intragastric administration,1~2 mL of abdominal aorta blood was taken and vena cava specimens were removed.The expression levels of RAGE and vascular cell adhesion molecule-1(VCAM-1)in serum of rats were detected by ELISA.The changes of blood vessel morphology were observed by HE staining.The changes of RAGE and VCAM-1 expression were detected by immunohistochemistry,WB and PCR.The results show that the intima injury of model group rats was serious,thrombosis was formed,and inflammatory cells infiltrated the vessel wall.The contents of RAGE and VCAM-1 in serum in model group were significantly higher than those in Chinese traditional medicine group and positive drug group.However,the expression of RAGE and VCAM-1 in the vascular tissue of rats in the Chinese traditional medicine group and the positive drug group were decreased.Compared with the sham operation group,the protein and mRNA expressions of RAGE and VCAM-1 in the model group were significantly increased,and after drug treatment,the protein and mRNA expressions of RAGE and VCAM-1 in the Chinese traditional medicine group and the positive drug group were significantly decreased.Therefore,excluding the effect of surgical trauma,antivenous thrombosis decoction may reduce the formation of venous thrombosis in DVT model rats by reducing the expression of RAGE and VCAM-1.
