摘 要:【目的】糖苷水解酶(GH)作为破坏植物细胞壁的重要酶类,在植物病原菌致病过程中发挥着重要作用。为了解GH蛋白之间的互作机制,对禾谷炭疽菌GH互作蛋白及其遗传关系进行研究。【方法】基于前期所获得的禾谷炭疽菌GH蛋白,通过STRING网络在线分析不同GH蛋白之间的互作关系,通过ProtComp v9.0在线分析GH蛋白的亚细胞定位,利用Clustal X和MEGA 11软件对GH蛋白进行多重比对和构建系统进化树,并选取互作关系多的蛋白通过GRAMM在线软件进行分子对接。【结果】禾谷炭疽菌中89个GH蛋白之间的互作表现为3大类的聚集关系,其中,Ⅰ类聚集了34个GH蛋白,Ⅱ类聚集了33个GH蛋白,Ⅲ类聚集了22个GH蛋白;85个GH蛋白定位于细胞外(分泌),仅4个GH蛋白定位于细胞核、内质网、细胞质膜。系统发育树分为4大类,有一些GH亚家族蛋白只存在于一类中,也有一些GH亚家族蛋白同时存在于两类及两类以上中;分子对接发现,互作蛋白之间均存在多个相互作用的氨基酸残基位点。【结论】禾谷炭疽菌GH蛋白之间有3种互作情况,即不同亚家族GH蛋白之间的互作、同一亚家族中不同GH蛋白之间的互作以及不存在互作关系;大部分GH蛋白定位于细胞外;大多数GH亚家族中不同蛋白亲缘关系较近,蛋白之间可通过氢键相互作用。【Objective】Glycoside hydrolases(GH),a group of enzymes that can destroy plant cell wall,play an important role in the invasion process of plant pathogens.In order to understand the interaction mechanism between GH proteins,the interaction and genetic relationship between GH proteins in Colletotrichum graminicola were studied.【Method】Based on previously obtained GH proteins of C.graminicola,the interaction relationship between different GH proteins was analyzed online through STRING network,and their subcellular localization was interpreted using ProtComp v9.0.Based on phylogenetic tree of the GH proteins constructed via Clustal X and MEGA 11 software,proteins with multiple interactions were selected for molecular docking through GRAMM online software.【Result】The interactions between 89 GH proteins in C.graminicola were clustered into 3 groups,in which GroupⅠ,ⅡandⅢcontained 34,33 and 22 GH proteins,respectively.Up to 85 GH proteins were located outside the cell(secreted),and the other 4 were located in the nucleus,endoplasmic reticulum,and cytoplasmic membrane.The 89 GH proteins were divided into 4 groups in the phylogenetic tree.Some GH subfamily proteins only existed in 1 group,while others coexisted in 2 or more groups.Molecular docking revealed multiple interactive amino acid residue sited between interactive proteins.【Conclusion】There are 3 types of interactions between GH proteins:interactions between GH proteins of different subclasses,interactions between different GH proteins in the same subclass,and no interaction relationship.Most GH proteins were located outside the cell,and different proteins within the same GH subfamily were closely related.Proteins can interact with each other through hydrogen bonding.
