摘  要:目的探讨过氧化物酶体增殖物激活受体γ辅激活因子1α(PGC-1α)、Ⅲ型纤连蛋白域蛋白5(FNDC5)在来曲唑诱导多囊卵巢综合征(PCOS)大鼠卵巢组织中的表达。方法将6周龄雌性SD大鼠随机分为对照组、PCOS组,每组各10只。PCOS组给予来曲唑溶液灌胃建立PCOS模型,对照组给予等体积羧甲基纤维素溶液灌胃,均1天1次,连续28 d。用化学发光法测定血清雄激素(T)、黄体生成素(LH)、卵泡雌激素(FSH)、空腹葡萄糖(FPG),计算LH/FSH;用ELISA法测定空腹血胰岛素(INS),计算胰岛素抵抗指数(HOMA-IR);HE染色后观察卵巢组织病理变化;免疫组化染色法检测卵巢组织PGC-1α、FNDC5蛋白表达。结果两组血清FPG比较差异无统计学意义(P>0.05),PCOS组血清T、LH、LH/FSH、INS、HOMA-IR较对照组高(P均<0.05),血清FSH水平较对照组低(P均<0.05)。对照组大鼠可见不同生长时期的卵泡及新鲜黄体,颗粒细胞整齐紧密排列、层数较多,可达6~8层;PCOS组可见多个未成熟的囊状卵泡及闭锁卵泡,颗粒细胞排列散乱不规则,层数较少。PGC-1α、FNDC5蛋白主要在卵巢组织颗粒细胞和卵丘细胞表达,呈现棕黄色,而卵泡膜细胞少量表达。与对照组比较,PCOS组卵巢组织PGC-1α蛋白表达低,FNDC5蛋白表达高,差异有统计学意义(P均<0.05)。结论PGC-1α在PCOS大鼠卵巢组织中低表达,FNDC5在PCOS大鼠卵巢组织中高表达。Objective To investigate the expression of peroxisome proliferator-activated receptor γ co-activator 1α(PGC-1α),and fibronectin type III domain-containing protein 5(FNDC5)in the ovarian tissues of rats with letrozole-induced polycystic ovary syndrome(PCOS).Methods Six-week-old female SD rats were randomly divided into the control group and PCOS group,with 10 rats in each group.Rats in the PCOS group were given letrozole solution by gavage to establish the PCOS models,and rats in the control group were given an equal volume of carboxymethyl cellulose solution by gavage,once a day,for 28 consecutive days.Chemiluminescence method was used to measure serum androgen(T),luteinizing hormone(LH),follicular estrogen(FSH),and fasting glucose(FPG);ELISA was used to measure fasting insulin(INS),and homeostatic model assessment for insulin resistance(HOMA-IR)was calculated;HE staining was used to observe the pathological changes of ovarian tissues;immunohistochemical staining and Western blotting were used to detect the protein expression levels of PGC-1αand FNDC5.Results There was no significant difference in serum FPG between the two groups(P>0.05),the serum levels of T,LH,LH/FSH,INS,HOMA-IR and FNDC5 in the PCOS group were higher than those in the control group(all P<0.05),and the serum levels of FSH and PGC-1αwere lower than those in the control group(all P<0.05).In the control group,follicles and fresh corpus luteum at different growth stages were visible,and the granulosa cells were neatly and tightly arranged,with a large number of layers,up to 6-8 layers.In the PCOS group,multiple immature cystic follicles and atresia follicles were seen,and the granulosa cells were arranged irregularly with a small number of layers.PGC-1αand FNDC5 proteins were mainly expressed in granulosa cells and cumulus cells,showing a brownish-yellow color,while follicular membrane cells were expressed in small amounts.Compared with the control group,the expression of PGC-1αprotein in the ovarian tissues was lower and the expression of F
