摘  要:以硫酸苯酚显色法测定白芨多糖的含量,超声法和温浸法探讨白芨多糖的提取方法,以正交实验和响应面法进行实验设计优选白芨多糖的最佳提取工艺。结果显示超声提取方法提取多糖含量跟水浴提取有显著性差异,正交实验通过回归分析,响应曲面通过直观等高线和三维立体图,综合评价了白芨多糖的最佳提取工艺为水提温度90℃,提取两次,每次1 h。多种实验设计比较能够全面分析,更切合实际确定白芨多糖提取工艺。The content of polysaccharides in Bletilla striata was determined by phenol sulfate.The extraction methods of Bletilla striata polysaccharide were studied by ultrasonic method and warm immersion method.The optimal extraction process of Bletilla striata polysaccharide was optimized by orthogonal experiment and response surface method.The content of polysaccharide extracted by ultrasonic extraction was significantly different from that by water bath extraction.The optimal extraction process of Bletilla striata polysaccharides was evaluated by orthogonal regression analysis,and the optimal extraction process was two times,1 h each,at the water extraction temperature of 90℃.Compared with various experimental designs,the extraction process of polysaccharides from Bletilla striata was determined more realistically.
