敷胸膏对呼吸道合胞病毒肺炎幼鼠肺组织病理改变及血清炎症因子的影响

摘　　要：目的观察敷胸膏对呼吸道合胞病毒肺炎幼鼠不同时相下的肺组织以及血清中白细胞介素-6(IL-6),IL-33和γ干扰素(IFN-γ)的影响。方法选取雄性幼龄SD大鼠54只,随机分为正常组、模型组和敷胸膏组3组,每组18只。呼吸道合胞病毒滴鼻感染幼龄鼠,在合胞病毒感染后的第3、5、7天取各组幼鼠血清、肺组织,苏木精-伊红染色观察病理改变,酶联免疫吸附试验(ELISA)法测定幼鼠肺血清中IL-6、IL-33及IFN-γ的变化。结果与正常组相比,模型组幼鼠IL-6、IL-33含量随着时间的变化逐渐升高(P<0.01),IFN-γ含量随着时间的变化逐渐降低(P<0.01)。模型组幼鼠肺泡壁有不同程度的增厚,出现间质性水肿以及炎细胞浸润。与模型组相比,敷胸膏组幼鼠IL-6、IL-33含量随着时间的变化逐渐降低(P<0.01),IFN-γ含量随着时间的变化逐渐升高(P<0.01)。敷胸膏能够缓解呼吸道合胞病毒肺炎幼鼠肺泡壁的增厚程度,降低间质性水肿,减少炎细胞浸润。结论敷胸膏能够降低呼吸道合胞病毒肺炎幼鼠血清中IL-6、IL-33的含量,升高血清中IFN-γ的含量,缓解呼吸道合胞病毒肺炎幼鼠肺组织的损伤程度。Objective To observe the effects of Fuxiong cream on the lung tissue and serum levels of interleukin-6(IL-6),interleukin-33(IL-33),and interferon-gamma(IFN-γ)in rats with respiratory syncytial virus(RSV)pneumonia at different time points.Methods Totally 54 young SD rats were randomly divided into three groups:normal group,model group,and Fuxiong cream group,with 18 rats in each group.RSV was intranasally administered to induce pneumonia in the rats,and serum and lung tissue samples were collected from each group at 3,5,and 7 days after infection.Hematoxylin-eosin staining(HE staining)was used to observe pathological changes,and ELISA was used to measure the levels of IL-6,IL-33,and IFN-γin mouse serum and lung tissue.Results Compared with the normal group,the levels of IL-6 and IL-33 in the model group gradually increased over time(P<0.01),while the level of IFN-γgradually decreased(P<0.01).The lung tissue of the model group rats showed varying degrees of thickening of the alveolar wall,interstitial edema,and inflammatory cell infiltration.Compared with the model group,the levels of IL-6 and IL-33 in the Fuxiong cream group gradually decreased over time(P<0.01),while the level of IFN-γgradually increased(P<0.01).Fuxiong Cream could alleviate the degree of alveolar wall thickening,reduce interstitial edema,and decrease inflammatory cell infiltration in rats with RSV pneumonia.Conclusion Fuxiong Cream can reduce the levels of IL-6 and IL-33 in the serum of rats with RSV pneumonia,increase the level of IFN-γ,and alleviate lung tissue damage in the young rats with RSV pneumonia.
