摘  要:目的:基于4D label-free定量蛋白质组学技术探究复发性流产(RSA)的发病机制。方法:纳入早期RSA患者和非医学原因行人工流产的受试者各8例,取受试者蜕膜组织进行蛋白质提取,胰酶酶解和液相色谱-质谱联用分析后得到原始数据,依据筛选标准获得差异表达蛋白,并进行交集映射,进行生物信息学分析。结果:两组共鉴定出5 063个蛋白质,共筛选出580个差异表达蛋白,其中有244个蛋白表达上调,336个蛋白表达下调。GO富集分析显示,差异蛋白主要集中在对外部刺激的反应、防御反应、生物黏附、细胞运动、细胞定位、细胞迁移、多细胞生物过程、细胞间通讯等生物过程。KEGG通路富集分析结果提示,差异蛋白参与了PI3K-AKT、p53信号通路、PPAR信号通路等。蛋白相互作用网络中筛选出FN1、FGF2、MPO、BGN、SPP1 5个核心蛋白。结论:RSA的发生机制可能涉及炎性反应、氧化应激、免疫调节、凝血调节、细胞黏附等生物过程及信号通路。利用4D label-free定量蛋白质组学技术和进一步的生物信息学分析,获得的差异蛋白及信号通路,对于揭示RSA的发病机制具有重要意义。Objective:To explore the pathogenesis of recurrent spontaneous abortion based on 4D label free quantitative proteomics technology.Methods:Eight patients with early recurrent spontaneous abortion and eight women who underwent non medical induced abortion were included.Protein extraction was performed on the decidual tissue of the subjects,followed by trypsin hydrolysis and liquid chromatography-mass spectrometry analysis to obtain raw data.Differential expression proteins were obtained based on screening criteria,and intersection mapping was performed for bioinformatics analysis.Results:A total of 5 063 proteins were identified in both groups,and a total of 580 differentially expressed proteins were screened,among which 244 proteins were up-regulated and 336 proteins were down-regulated.GO enrichment analysis showed that differential proteins were mainly concentrated in biological processes such as response to external stimulus,defense response,biological adhesion,cell motility,localization of cell,cell migration,multicellular organismal process,cell communication.The results of KEGG pathway enrichment analysis showed that differential proteins participated in PI3K-AKT,p53 signaling pathway and PPAR signaling pathway.Five core proteins,including FN1,FGF2,MPO,BGN and SPP1,were screened out in the protein interaction network.Conclusion:The mechanism of recurrent spontaneous abortion may involve biological processes and signaling pathways such as inflammation,oxidative stress,immunomodulation,coagulation regulation,and cell adhesion.The differential proteins and signaling pathways obtained by using 4D label-free quantitative proteomics technology and further bioinformatics analysis are of great significance for revealing the pathogenesis of recurrent miscarriage.
