摘 要:铁死亡是一种铁依赖的程序性细胞死亡,细胞内铁过载和脂质活性氧蓄积对铁死亡的发生发展起核心促进作用,任何影响铁代谢和氧化还原系统平衡的因素都可能诱发加重铁死亡。铁死亡参与急性肾损伤、糖尿病肾病、肾间质纤维化等诸多肾脏病的病理过程,且抑制铁死亡具有肾脏保护作用。该文系统综述铁死亡的病理生理机制及其在肾脏病中的研究进展,探讨靶向铁死亡在肾脏病治疗中的应用前景。Ferroptosis is an iron-dependent programmed cell death.Iron overload and lipid reactive oxygen accumulation play a core role in the occurrence and development of ferroptosis.Any factors affecting the balance of iron metabolism and redox system may induce and aggravate ferroptosis.Ferroptosis is involved in the pathological process of acute kidney injury,diabetic nephropathy,renal interstitial fibrosis and some other kidney diseases,and inhibiting ferroptosis has potential renoprotective benefits.This article reviewed the pathophysiological mechanism of ferroptosis and its research progress in renal diseases,and discussed the application prospect of targeted ferroptosis in the treatment of renal diseases.
