摘  要:目的 利用网络药理学和生信分析来探究委陵菜治疗胃癌的有效机理。方法 采用的数据库信息网络平台属于中药系统药理学的范围(the traditional Chinese medicine database analysis platform, TCMSP),并在其中建立了特殊的筛查标准,以确保委陵菜中所含有的有效成分被准确检测出来。经过研究发现委陵菜的主要活性成分的靶点可以在多个数据集中得到,包括Swiss、SEA、TCMSP、PharmMapper等。从GEO(取3个GSE文件)数据库获取胃癌的主要作用靶点,将以上4个数据库预测的药物靶点与3个GSE数据集预测的胃癌靶点取交集得到委陵菜与胃癌的共同靶点;通过STRING数据库对两者的共同靶点进行蛋白互作(protein-protein interaction network, PPI)网络分析搭建了PPI模型,探索委陵菜和胃癌之间的关联性。通过Metascape平台对委陵菜与胃癌的共同靶点进行GO与KEGG富集分析,同时采用微生信在线生物信息学分析、可视化云平台进行可视化分析。结果 利用TCMSP平台筛选得委陵菜6种有效活性成分及748个作用靶点,3个GSE数据集得到胃癌靶点833个,取交集得到委陵菜与胃癌的49个共同靶点;委陵菜治疗胃癌的有效活性成分分别为丁子香萜、鞣花酸、异鼠李素、槲皮素、山柰酚、β-谷甾醇等;对应的靶点分别为CXCL8、SERPINE1、SPP1、CYP2C19、GSTA1等;主要通过PI3K-Akt、MAPK、P53等信号通路发挥作用。结论 委陵菜治疗胃癌的特点主要有成分复杂、靶点丰富、通路繁多,结合网络药理学理论,再通过生信分析预测到委陵菜可能通过PI3K-Akt、MAPK、P53等相关通路能够产生治疗胃癌的效果,从理论层面为胃癌的相关探索提供参考。Objective To explore the effective mechanisms of potentilla chinensis in treating gastric cancer utilizing network pharmacology and bioinformatics analysis.Methods The Traditional Chinese Medicine Database Analysis Platform(TCMSP)was employed within specific screening criteria to accurately detect the effective components in potentilla chinensis.The main active ingredient targets of potentilla chinensis were identified across various datasets including Swiss,SEA,TCMSP,PharmMapper,etc.The primary action targets of gastric cancer were obtained from three GSE files in the GEO database.The intersection of the drug targets predicted by the four databases and the gastric cancer targets predicted by the three GSE datasets yielded the common targets of potentilla chinensis and gastric cancer.A protein-protein interaction(PPI)network analysis was constructed using the STRING database to explore the relationship between potentilla chinensis and gastric cancer through their common targets.The common targets of potentilla chinensis and gastric cancer were subjected to GO and KEGG enrichment analysis using the Metascape platform,alongside visualization analysis through bioinformatics and visualization cloud platforms.Results 6 effective active ingredients and 748 target points were identified in potentilla chinensis through the TCMSP platform.833 gastric cancer targets were obtained from three GSE datasets,resulting in 49 common targets shared by potentilla chinensis and gastric cancer.The active components of potentilla in the treatment of gastric cancer were eugenol,ellagic acid,isorhamnetin,quercetin,kaempferol,β-sitosterol,etc.Corresponding targets included CXCL8,SERPINE1,SPP1,CYP2C19,GSTA1 and so on,acting primarily through pathways like PI3K-Akt,MAPK,P53,etc.Conclusion The characteristics of potentilla chinensis in treating gastric cancer involve complexity in composition,rich target points,and diverse pathways.Applying network pharmacology theory and bioinformatics analysis,it is predicted that potentilla chinensis mi
