摘 要:三维超分辨成像技术在深入探究细胞内亚细微结构方面有广泛的应用,但是其研发与应用需要准确的成像校准工作。柱透镜是实现三维超分辨成像的关键光学工具之一,探究光路变化尤其是柱透镜、相机成像面和显微镜镜筒透镜之间的相对距离对于实现准确的三维超分辨成像具有重要的意义。基于此,聚焦于影响三维超分辨成像校准的主要因素,并利用校准完备的三维超分辨荧光显微成像系统,实现对细胞膜三维结构的清晰成像,进一步对荧光分子在细胞膜结构中横向(x轴和y轴方向)及轴向(z轴方向)扩散速率进行解析,从而有望为三维超分辨成像技术提供详细与广泛的应用推广。Three dimensional super-resolution imaging technique has been widely used in intracellular imaging of sub-micrometer structures,but its developments and applications require a precise calibration.The cylindrical lens is one of the key optics in three dimensional super-resolution imaging.Study of how the distance between cylindrical lens,camera imaging plane,and the microscope tube lens remains to be a significant task on three dimensional super-resolution imaging.Based on these,this paper focused on the investigations of several key elements that determine the calibration of three dimensional super-resolution imaging.Through careful calibration on how camera-lens distance impacted the imaging system,this paper achieved super-resolution imaging of intracellular membrane structures on three dimensions,and spatially mapped the diffusion rates along x-y direction and z axis at the nanoscale.Taken together,these results provide a detailed protocol on the calibration of three dimensional super-resolution imaging system,and pave the way for its wider application in intracellular imaging.
