摘 要:目的研究建立仙芎骨康颗粒的最佳提取工艺。方法采用正交试验法考察提取次数、溶剂用量和提取时间等因素对提取工艺的影响,以淫羊藿苷、朝藿定C、宝藿苷I、阿魏酸、齐墩果酸、桂皮醛等6成分的转移率为评价指标,层次分析法确定各成分的权重系数(权重系数分别为淫羊藿苷0.35、朝藿定C 0.14、宝藿苷I 0.09、阿魏酸0.2、齐墩果酸0.13、桂皮醛0.09),采用多波长切换-梯度洗脱技术建立仙芎骨康药液6成分的HPLC含量测定方法;以多指标综合加权评价法确定仙芎骨康颗粒的最佳提取工艺,并进行3批验证试验。结果影响提取效率的主次因素为:提取时间>提取次数>固液比例。仙芎骨康颗粒的最佳提取工艺为:以10倍量50%乙醇回流提取2次,每次提取1 h。结论优选得到的仙芎骨康提取工艺能较完全地提取药效成分,方法稳定重现,适用于工业化生产质量控制。Objective To study and establish the best extraction process of Xianxiong Gukang particles.Methods The orthogonal test was used to investigate the effects of extraction times,solid-liquid ratio and extraction time on the extraction process.Taking icariin,epimedin C,baohuoside I,ferulic acid,oleanolic acid,cinnamic aldehyde transfer rate as evaluation index,and the analytic hierarchy process to determine the components weight coefficient(weight coefficients were 0.35,0.14,0.09,0.2,0.13,0.09),and adopts multi-wavelength switching and gradient elution technology to establish a HPLC method for the determination of the above six components in Xianxiong Gukang liquid medicine;The multi-index comprehensive weighted evaluation method was used to determine the best extraction process,and three batches of validation tests were carried out.Results The factors influencing the efficiency were,in order,extraction time,extraction times and solid-to-liquid ratio.The best extraction process was that extracted by 10 times of 50%ethanol twice and 60 mins per time.Conclusion The optimized extraction process of Xianxiong Gukang can completely extract the effective components,and the method is stable and reproducible,which is suitable for quality control of industrial production.
