摘  要:为了研究环氧二十碳三烯酸(epoxyeicosatrienoic acids, EET)对肥胖大鼠白色脂肪棕色化的影响,试验将32只4周龄雄性SD大鼠随机分为对照组(n=8)和高脂组(n=24),高脂组大鼠连续饲喂高脂饲料15周,将建模成功的肥胖大鼠再随机分为模型对照组和EET组(11,12-EET组和14,15-EET组),每组8只。EET组大鼠每天按体重30μg/kg皮下分别注射11,12-EET和14,15-EET,对照组和模型对照组皮下注射同等剂量生理盐水,每周测量体重。10 d后采用ELISA、实时荧光定量PCR方法检测大鼠血脂含量、口服糖耐量及脂肪组织中棕色化相关指标解偶联蛋白1(uncoupling protein 1,UCP-1)、血管内皮生长因子(vascular endothelial growth factors, VEGF)和过氧化物酶体增殖激活受体γ辅激活因子1α(peroxisome prolifertor-activated receptor-γ coactivator-1α,PGC-1α)基因的相对表达量。结果表明:高脂饲料饲养15周后,高脂组体重显著高于对照组(P<0.05)。与模型对照组相比,11,12-EET组和14,15-EET组大鼠在口服葡萄糖2 h后,血糖浓度缓慢恢复至正常值;11,12-EET组和14,15-EET组大鼠血清三酰甘油(TG)含量显著降低(P<0.05);11,12-EET组大鼠腹股沟脂肪组织中UCP-1基因相对表达量显著升高(P<0.05),14,15-EET组大鼠肩胛骨脂肪组织中UCP-1基因相对表达量显著升高(P<0.05);14,15-EET组和11,12-EET组大鼠腹股沟和肩胛骨脂肪组织中VEGF基因相对表达量均显著降低(P<0.05);14,15-EET组和11,12-EET组大鼠肩胛骨脂肪组织中PGC-1α基因相对表达量显著降低(P<0.05),腹股沟脂肪组织中PGC-1α基因相对表达量显著升高(P<0.05)。说明EET可能通过改善糖脂代谢促进肥胖大鼠白色脂肪棕色化过程。The aim of the present study was to investigate the effects of epoxyeicosatrienoic acids(EET)on the browning of white adipocytes in obese rats.Thirty-two 4-week-old male SD rats were randomly divided into control group(n=8)and high fat group(n=24).The rats in the high-fat group were fed high-fat diet continuously for 15 weeks.The successfully modeled obese rats were then randomly divided into model control group and EET group(11,12-EET and 14,15-EET groups),with 8 rats in each group.Rats in EET group were injected subcutaneously with 11,12-EET and 14,15-EET at the weight of 30μg/kg,respectively,and the control group and model group were injected subcutaneously with the same dose of normal saline,and their weight was measured weekly.After 10 days,oral glucose tolerance test was performed,and the blood lipid content and browning-related indexes in adipose tissue were detected by ELISA and real-time quantitative PCR to detect the blood lipid content and browning related indexes in adipose tissue uncoupling protein 1(UCP-1),vascular endothelial growth factors(VECF)and the relative expression of peroxisome proliferation activating receptor coactivator lα(PCC-lα).The results showed that the body weight of high fat group was significantly higher than that of control group after feeding high fat diet for 15 weeks(P<0.05).After oral administration of glucose for 2 h,the blood glucose concentration of 11,12-EET and 14,15-EET group returned to normal value slowly.Compared with model control group,serum triglyceride(TG)content in 11,12-EET and 14,15-EET groups was significantly decreased(P<0.05).The relative expression level of UCP-1 gene in groin adipose tissue of 11,12-EET group was significantly increased(P<0.05),and the relative expression level of UCP-1 gene in scapular adipose tissue of 14,15-EET group was significantly increased(P<0.05).In the 14,15-EET and 11,12-EET groups,the relative expression of the VECF gene in inguinal and scapular adipose in rats was significantly decreased(P<0.05).The relative expression o
