摘 要:目的探讨和厚朴酚对非酒精性脂肪性肝病(non-alcoholic fatty liver disease,NAFLD)大鼠模型的改善作用及相应的调控机制。方法将72只SD雄性大鼠分为正常组和造模组,正常组给予普通饲料,造模组给予高脂饲料,喂养8周后各取6只验模。成功建立NAFLD模型后,将其分为正常组、模型组、和厚朴酚低剂量组、和厚朴酚中剂量组、和厚朴酚高剂量组以及多烯磷脂酰胆碱组,每组10只。各组给予相应的生理盐水或药物灌胃6周,14周末取材后采用生化法检测各组大鼠空腹血糖(FBG)、血清胰岛素(INS)、肝功能(ALT、AST)、血脂(TC、TG、HDL-C、LDL-C)、氧化应激指标(MDA、SOD、T-AOC)、炎性细胞因子(IL-8、TNF-α)水平;采用HE染色观察各组大鼠肝组织病理形态学改变情况;采用免疫荧光法和Western Blotting检测各组大鼠肝组织PPARα、ACOX1和CPT1A的蛋白表达情况。结果HE染色显示,与正常组相比,模型组出现大量脂肪空泡,和厚朴酚低、中、高3个剂量以及多烯磷脂酰胆碱能够显著减少脂肪空泡。体质量称量结果显示,各治疗组能够显著降低NAFLD大鼠体质量水平;生化指标结果显示,各治疗组能够显著降低NAFLD大鼠血糖、胰岛素水平,以及改善肝功能(ALT、AST)、血脂(TC、TG、HDL-C、LDL-C)、氧化应激指标(MDA、SOD、T-AOC)、炎性细胞因子(IL-8、TNF-α)水平。Western Blotting和免疫荧光结果显示,模型组PPARα、ACOX1和CPT1A表达显著低于正常组,而各给药组能够显著逆转这一效应。结论和厚朴酚能够调控脂肪酸β氧化,改善脂代谢,降低氧化应激和炎性反应,从而保护NAFLD大鼠。Objective To explore the improvement effect and corresponding regulatory mechanism of honokiol on a rat model of non-alcoholic fatty liver disease(NAFLD).Methods 72 SD male rats were divided into a normal group and a model group.The normal group was given regular feed,while the model group was given high-fat feed.After 8 weeks of feeding,6 rats were taken from each group for model testing.After the successful establishment of NAFLD model,they were divided into normal group,model group,and honokiol low dose group,and honokiol medium dose group,and honokiol high dose group,and polyenyl phosphatidylcholine group.Each group was given corresponding saline or drug gavage for 6 weeks,and the levels of fasting blood glucose(FBG),serum insulin(INS),liver function(ALT,AST),blood lipids(TC,TG,HDL-C,LDL-C),oxidative stress indexes(MDA,SOD,T-AOC),inflammatory cytokines(IL-8,TNF-α)were measured by biochemical methods after sampling at the end of 14 weeks in each group;HE staining was used to observe the histomorphological changes in the liver of each group of rats;The protein expression of PPARα,ACOX1 and CPT1A in the liver tissues of rats in each group was detected by immunofluorescence and Western Blotting.Results HE staining showed that a large number of fat vacuoles appeared in the model group compared with the normal group,and that the three doses of chondroitol low,medium and high as well as polyenyl phosphatidylcholine could significantly reduce the fat vacuoles.The results of body weight weighing showed that each treatment group was able to significantly reduce the body weight level of NAFLD rats;The results of biochemical indexes showed that each treatment group was able to significantly reduce blood glucose and insulin levels,as well as reduce serum liver function(ALT,AST),blood lipids(TC,TG,HDL-C,LDL-C),oxidative stress indexes(MDA,SOD,T-AOC),inflammatory cytokines(IL-8,TNF-α)levels,etc.in NAFLD rats.Western Blotting and immunofluorescence results showed that PPARα,ACOX1 and CPT1A expressions were significantly
