乳酸脱氢酶法和四唑盐比色法对纳米银抗菌凝胶细胞毒性评价的比较研究

摘　　要：目的:采用两种体外细胞毒性方法评价纳米银抗菌凝胶的细胞毒性,旨在评估两种方法在纳米银抗菌凝胶毒性评价中的适宜性及优缺点,为该产品的细胞毒性研究提供技术支持。方法:采用乳酸脱氢酶(LDH)试验和四唑盐比色法(MTT试验)对抗菌凝胶进行细胞毒性评价。结果:MTT试验结果显示抗菌凝胶100%浸提液(0.1g/mL)和70%、60%、50%、40%、20%系列稀释液细胞相对存活率分别为:1%,2%,1%,27%,89%,96%;LDH试验结果显示抗菌凝胶100%浸提液(0.1g/mL)和70%、60%、50%、40%、20%稀释液的LDH总释放率为:2%,3%,10%,55%,28%,16%。MTT试验的IC50为0.048g/mL,抗菌凝胶IDH试验的LC50为0.051g/mL。结论:两种方法测得的细胞毒性随着试验样品浓度的降低均降低,两者的趋势一致,且其IC50浓度接近,两种方法从不同角度阐明了该产品的毒性,为临床合理用药提供数据支持。Objective:Two in vitro cytotoxicity methods were used to detect the cytotoxicity of nanosilver antibacterial gel.The purpose is to evaluate the suitability and disadvantages of the two methods and to provide technical support for the cytotoxicity study.Methods:Cytotoxicity tests were conducted by LDH test and MTT test.Results:MTT test showed the Relative growth rate of 0.1g/mL of test article extract and 70%extract,60%extract,50%extract,40%extract,20%extract were 1%,2%,1%,27%,89%and 96%respectively;LDH test showed the total release rate of 0.1g/mL of test article extract and 70%extract,60%extract,50%extract,40%extract,20%extract were 2%,3%,10%,55%,28%and16%respectively.The IC50 of test article detected from MTT assay was 0.048g/mL and the IC50 of that detected from LDH assay was 0.051g/mL.Conclusion:Cytotoxicity in two tests decreases with the decrease of test article extract concentration,two methods show the same trend,and their IC50 concentration is close.Two methods clarify the toxicity of test article from different angles and provide data support for clinical drug use.
