摘  要:为了解胰岛素样生长因子1受体(IGF1R)基因外显子多态性与昆明犬体重和体尺性状的关系,从而为优质昆明犬的培育寻找可靠的分子选择标记,试验采用PCR-单链构象多态性(PCR-SSCP)技术对昆明犬IGF1R基因外显子区域单核苷酸多态性(single nucleotide polymorphism, SNP)位点进行筛选,通过测序分析其突变情况,统计SNP位点的优势基因型、优势等位基因、多态信息含量(polymorphism information content, PIC)、纯合度(homozygosity, Ho)、杂合度(heterozygosity, He)、有效等位基因数(number of effective alleles, Ne)、Hardy-Weinberg平衡状态,并采用一般线性模型分析SNP位点与昆明犬体重和体尺性状的关联性。结果表明:在昆明犬IGF1R基因外显子中仅识别到1个SNP位点,该位点位于外显子8的第1 773位核苷酸处,发生了C→T的点突变,但氨基酸未变,命名为C1773T位点。C1773T位点在昆明犬群体中共存在两种基因型,分别为CC纯合型和CT杂合型,其中优势基因型为CC纯合型,优势等位基因为C,表现为低度多态(PIC<0.25),Ho为0.97,He为0.03,Ne为1.03,在昆明犬群体中处于Hardy-Weinberg平衡状态。昆明犬IGF1R基因C1773T位点CT杂合型个体胸宽显著高于CC纯合型个体(P<0.05),其他体重和体尺性状指标均差异不显著(P>0.05)。说明IGF1R基因C1773T位点对昆明犬胸宽具有一定影响,可作为昆明犬品种培育的潜在分子选择标记。In order to understand the relationship between exon polymorphisms of the insulin-like growth factor 1 receptor(IGF1R)gene and weight and body size traits in Kunming dogs,so as to find reliable molecular selection markers for the breeding of high-quality Kunming dogs,in the experiment,the single nucleotide polymorphism(SNP)loci in the exonic region of Kunming dog IGF1R gene were screened by PCRsingle strand conformation polymorphism(PCR-SSCP).The mutation status was analyzed by sequencing,and dominant genotypes,dominant alleles,polymorphic information content(PIC)homozygosity(Ho),heterozygosity(He),number of effective alleles(Ne)and Hardy-Weinberg equilibrium status of SNP loci were counted.Finally,general linear model was used to analyze the association between SNP loci and body weight and body size traits in Kunming dogs.The results showed that only one SNP locus was recognized in the exon of Kunming dog IGF1R gene;the site was located at nucleotide 1773 of exon 8,with a point mutation of C→T but no change in amino acid,and was named C1773T locus.There were two genotypes of the C1773T locus in the Kunming dog population,CC homozygotic type and CT heterozygous type,in which the dominant genotype was CC homozygotic type and the dominant allele was C,which showed a low degree of polymorphism(PIC<0.25);Ho was 0.97;He was 0.03,and Ne was 1.03,which was in Hardy-Weinberg equilibrium in the Kunming dog population.The chest width of CC homozygotic type individuals was significantly higher than that of CT heterozygous individuals in the C1773T locus of the IGF1R gene in Kunming dogs(P<0.05);the differences in other physical traits were not significant(P>0.05).The results indicated that the C1773T locus of the IGF1R gene had a certain effect on the chest width of Kunming dogs,which could be used as a potential molecular selection marker for Kunming dog breeding.
