芫根薤白压片糖果对小鼠抗缺氧和学习记忆能力的影响

摘  要:目的:探讨芫根薤白压片糖果对小鼠抗缺氧和学习记忆能力的影响。方法:首先,建立常压缺氧模型、急性脑缺氧模型、亚硝酸钠中毒致缺氧模型和异丙肾上腺素致缺氧模型。观察小鼠的存活时间,采用ELISA法检测心脏和脑组织的超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)含量。其次,小鼠腹腔注射东莨菪碱(2 mg·kg^(-1)),通过跳台实验观察小鼠的记忆能力。采用ELISA法检测脑组织的乙酰胆碱(acetylcholine,Ach)、乙酰胆碱酯酶(acetylcholinesterase,AchE)和胆碱乙酰转移酶(choline acetyltransferase,ChAT)含量。结果:在缺氧模型中,芫根薤白压片糖果可延长小鼠的存活时间,并增加心脏和脑组织GSH-Px、SOD含量(P<0.05)。在记忆能力实验中,芫根薤白压片糖果能减少小鼠的跳台次数,并增加脑组织Ach、ChAT含量,降低脑组织AchE含量。结论:芫根薤白压片糖果可能通过增加心脏和脑组织的抗氧化酶含量从而提高小鼠抗缺氧能力,同时通过调节脑内神经递质含量从而增强小鼠的学习记忆能力。Objective:To investigate the effects of Yuangen Xiebai Tablet Candy on anti-hypoxia as well as learning and memory ability of mice.Methods:Firstly,normobaric hypoxia models,acute cerebral hypoxia models,hypoxia models induced by sodium nitrite poisoning and hypoxia models induced by isoproterenol were established.The survival time of mice was observed,and the contents of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in heart and brain tissues were detected by ELISA.Secondly,the mice were intraperitoneally injected with scopolamine(2 mg·kg^(-1)),and the memory ability of which was observed through the platform experiment.The contents of acetylcholine(Ach),acetylcholinesterase(AchE)and choline acetyltransferase(ChAT)in brain tissue were detected by ELISA.Results:In the hypoxia model,the Yuangen Xiebai Tablet Candy treatment prolonged the survival time of the mice and increased the contents of GSH-Px and SOD in the heart and brain tissues significantly(P<0.05).In the memory ability test,Yuangen Xiebai Tablet Candy can reduce the number of jumps,increase the content of Ach and ChAT while reduce the content of AchE in brain tissue.Conclusion:Yuangen Xiebai Tablet Candy can improve the anti-hypoxia ability of mice by increasing the content of antioxidant enzymes in the heart and brain tissues and enhance the learning and memory abilities of mice by regulating the content of neurotransmitters in the brain.
