摘 要:为探究γ射线辐照灭活对牛血清品质的影响,以胎牛血清、新生牛血清为材料,研究8、10、11、12、13kGy 5个不同辐照剂量对牛血清中大肠埃希菌噬菌体的影响,以及12、20、25、30、35、40 kGy 6个不同辐照剂量对牛血清中主要成分含量、细胞增殖的影响。结果表明,≥12 kGy剂量的γ射线辐照能灭活牛血清中的病原微生物;辐照对除血红蛋白外的牛血清主要成分含量无显著影响,也不会降低内毒素含量;牛血清经≤35 kGy辐照剂量处理后,其Sp2/0-Agl4细胞的倍增时间和克隆率、Mv1 Lu细胞倍增时间均符合《中华人民共和国药典2020年版四部》新生牛血清标准。本研究可为γ射线辐照灭活在牛血清中的应用提供理论依据与技术支持。The aim of this study was to explore the influence of γ-ray irradiation inactivation on the quality of bovine serum.The fetal bovine serum and newborn bovine serum were used as samples.The effects of five different irradiation doses of 8,10,11,12,and 13 kGy on coliform bacteriophages in bovine serum were studied.The effects of six different irradiation doses of 12,20,25,30,35,and 40 kGy on the content of main ingredients and cell proliferation in bovine serum were studied.The results indicate that aγ-irradiation dose of≥12 kGy could inactivate pathogenic microorganisms without significant impact on the content of major components in bovine serum except hemoglobin,nor did it reduce the endotoxin content.The doubling time and cloning rate of Sp2/0-Agl4 cells,as well as the doubling time of Mv1 Lu cells of bovine serum treated with doses of≤35 kGy were in line with the standards for newborn bovine serum in the Pharmacopoeia of the People’s Republic of China,2020 Edition,Part Four.This study provides theoretical basis and technical support for the application of bovine serum γ-irradiation inactivation.
