摘 要:以‘豫菊花2号’药食两用菊花茎段为材料,经过组织培养和脱毒处理,建立‘豫菊花2号’菊花种苗快速繁育体系,确定株高和地茎为分级的量化指标。结果表明:‘豫菊花2号’茎段的最佳灭菌方案为84消毒液(含氯量3.4%~4.6%)浸泡25 min;茎尖诱导最佳激素配比为6-BA 1.5 mg/L+NAA 0.1 mg/L;通过聚类分析将‘豫菊花2号’脱毒苗分为Ⅰ级、Ⅱ级、Ⅲ级苗,分级标准:Ⅰ级脱毒苗的等级标准为株高不小于7.5 cm、地茎不小于1.75 mm,共11株;Ⅱ级脱毒苗的等级标准为株高在5.0~7.5 cm之间、地茎在1.50~1.75 mm之间,共52株;Ⅲ级脱毒苗的等级标准为株高在2.5~5.0 cm之间、地茎小于1.50 mm,共27株,符合正态分布。本研究为‘豫菊花2号’品种推广生产提供参考依据。Taking the stem of'Yujuhua 2 Hao'as the test material,tissue culture and detoxification treatment were studied to establish a rapid breeding system for'Yujuhua 2 Hao'chrysanthemum seedlings.The plant height and ground stem were determined as quantitative indicators for the classification.The results showed that the best sterilization plan for the stem of'Yujuhua 2 Hao'was soaked in 84 disinfectant solution(3.4%~4.6%chlorine)for 25 min;the best stem tip inducing hormone ratio was 6-BA 1.5 mg/L+NAA 0.1 mg/L.Through cluster analysis,'Yujuhua 2 Hao'tissue cultured seedlings were divided into levelⅠ,levelⅡand levelⅢ.The grading criteria:the grade standard for Grade I tissue culture seedlings was not less than 7.5 cm in plant height and ground stem was not less than 1.75 mm,a total of 11 plants.The grade standard for GradeⅡwas between 5.0~7.5 cm in plant height and 1.50~1.75 mm in stem,a total of 52 plants.The grade standard for GradeⅢtissue culture seedlings was 2.5~5.0 cm in plant height and less than 1.50 mm in stem,a total of 27 plants,conform to normal distribution.This study provides a reference basis for the promotion and production of'Yujuhua 2'variety.
