白鲜皮中白鲜碱的提取工艺优化研究

摘　　要：通过单因素、正交实验设计优化白鲜皮中白鲜碱的提取工艺,白鲜碱作为对照品,制作标准曲线,采用高效液相色谱仪测定白鲜碱的含量。结果显示以乙醇作为溶剂时,白鲜碱的最佳提取工艺是乙醇浓度为90%,提取时间为1.5 h,料液比为1∶4,温度为70℃,在最佳提取工艺下,白鲜皮醇提取液中白鲜碱的含量为0.94 mg/g。标准曲线回归方程为y=215315x+740.97,R^(2)=0.999,在0.0005~0.02 mg/mL范围内呈良好线性关系。本工艺方法操作简便,易实现,为下一步提取白鲜碱进一步活性研究提供依据。The extraction process of dictamnine from cortex was optimized by one-way and orthogonal experimental design,and leucovorin was used as a control to make a standard curve,and the content of dictamnine was determined by HPLC.The results showed that when ethanol was used as the solvent,the optimal extraction process of cortex dictamnine was ethanol concentration of 90%,extraction time of 1.5 h,material-liquid ratio of 1∶4,and temperature of 70℃.under the optimal extraction process,the content of leucovorin in the alcoholic extract of dictamnine was 0.94 mg/g.The regression equation of the standard curve was y=215315x+740.97,R^(2)=0.999,and it was found to be in the range of 0.0005~0.02 mg/mL range showed good linear relationship.This process method is simple and easy to realize,and provides the basis for further activity study of the next extraction of dictamnine.
