摘  要:目的建立一种测定黄芪桂枝五物汤和芍药水提物中芍药苷和芍药内酯苷含量的超快速液相色谱法。方法采用ShimPack XR-ODS色谱柱,以0.1%磷酸水(A)和乙腈(B)作为流动相,梯度洗脱(0~5 min,10%-20%B;5~10 min,20%-25%B),平衡时间为5 min,流动相的流速为0.4 mL/min,色谱柱的温度设定为35℃,紫外检测器的波长设定为230 nm,进样量设定为5μL。结果芍药苷和芍药内酯苷的线性范围均为0.5~20 mg/L,所建立的分析方法具有较好的线性关系、精密度、稳定性和回收率。黄芪桂枝五物汤和芍药水提物中芍药苷的含量分别为2.75~3.84 g/L和4.70~4.99 g/L,芍药内酯苷的含量分别为0.632~0.928g/L和0.729~0.870 g/L。结论芍药苷在黄芪桂枝五物汤中比在芍药水提物中的含量少,表明其它药材成分对芍药苷的提取具有抑制作用。芍药内酯苷在黄芪桂枝五物汤中和在芍药水提物中含量无明显差异。本方法分析时间短、稳定性好,可用于黄芪桂枝五物汤的质量控制和配伍规律研究。Objective To establish an ultra-fast liquid chromatography method for the determination of paeoniflorin and albiflorin in Huangqi Guizhi Wuwu Decoction(HGWWD)and Paeoniae Radix Alba water extract.Methods Shim-Pack XR-ODS chromatographic column was used,0.1%phosphoric acid water(A)and acetonitrile(B)were adopted as mobile phase,and gradient elution program(0-5 min,10%-20%B;5-10 min,20%-25%B)was applied for good separation.The equilibrium time was 5 min,the flow rate of the mobile phase was 0.4 mL/min,the temperature of the chromatographic column was set to 35℃,the wavelength of the UV detector was set to 230 nm,and the injection volume was set to 5μL.Results The linear range of paeoniflorin and albiflorin was 0.5-20 mg/L.The developed analytical method has excellent linear relationship,good precision,good stability and stable recovery.The contents of paeoniflorin were 2.75-3.84 g/L and 4.70-4.99 g/L,and the contents of albiflorin were 0.632-0.928 g/L and 0.729-0.870 g/L in HGWWD and Paeoniae Radix Alba water extract.Conclusion The content of paeoniflorin in HGWWD was less than that in the water extract of Paeoniae Radix Alba,indicating that other medicinal ingredients have an inhibitory effect on the extraction of paeoniflorin.There was no significant difference in the content of albiflorin in HGWWD and in the water extract of Paeoniae Radix Alba.This method has the advantages of short analysis time and good stability,and can be used for quality control and compatibility study of HGWWD.
