加味当归补血汤对正常小鼠免疫功能的作用研究

摘　　要：[目的]探究加味当归补血汤对正常小鼠免疫功能的影响。[方法]将120只BALB/c小鼠随机分成3组,即免疫Ⅰ组、免疫Ⅱ组、免疫Ⅲ组,每个免疫组小鼠按体质量随机分为正常对照组、加味当归补血汤1.4 g·kg^(-1)组、加味当归补血汤1.9 g·kg^(-1)组、加味当归补血汤2.3 g·kg^(-1)组。连续灌胃30 d后,检测免疫Ⅰ组小鼠行为学指标,脾脏和胸腺系数,脾脏T淋巴细胞增殖能力,NK细胞活性,血常规指标,血清免疫球蛋白M(immunoglobulin M,IgM)和免疫球蛋白G(immunoglobulin G,IgG)含量,外周血和脾脏中T、B淋巴细胞比例;检测免疫Ⅱ组小鼠溶血空斑数、半数溶血值;检测免疫Ⅲ组小鼠腹腔巨噬细胞吞噬鸡红细胞的吞噬功能和脾脏中CD4、CD8表达量。[结果]与正常对照组比较,加味当归补血汤3个剂量均可明显升高小鼠自主活动次数和抓力,脾脏系数和胸腺系数(P<0.05,P<0.01);增强脾淋巴细胞增殖能力和NK细胞活性(P<0.01);增加溶血空斑数、半数溶血值、腹腔巨噬细胞对鸡红细胞的吞噬率和吞噬指数(P<0.01);升高全血淋巴细胞(lymphocyte,LYMPH)、白细胞(white blood cell,WBC)、红细胞(red blood cell,RBC)数量,血清中IgM、IgG含量(P<0.05,P<0.01);降低外周血和脾脏CD3^(+)T、CD8^(+)T细胞占比,减少CD8阳性表达量(P<0.05,P<0.01);升高外周血CD19+B细胞占比,外周血和脾脏CD4+/CD8^(+)比值,以及脾脏CD4/CD8阳性表达比值(P<0.05,P<0.01)。[结论]加味当归补血汤能增强特异性免疫和非特异免疫,有助于增强免疫力。[Objective]To investigate the implication of modified Duanggui Buxue Decoction on the immune function of normal mice.[Methods]A total of 120 BALB/c mice were stochastically divided into three groups,immunization group I,immunization group I and immunization group II.According to body weight,each immune group was randomly divided into 4 groups:normal control group,modified Duanggui Buxue Decoction 1.4 g·kg^(-1 )group,modified Duanggui Buxue Decoction 1.9 g•kg^(-1 )group,modified Duanggui Buxue Decoction 2.3 g·kg^(-1 ) group.After being gavaged for 30 d,for immune group I:test the behaviors,spleen index,thymus index,the proliferative capacity of splenic T lymphocytes,NK cell activities,blood indices,serum levels of immunoglobulin M(IgM)and immunoglobulin G(IgG),the ratios of T and B lymphocytes in the peripheral blood and spleen.Immune group II:number of hemolytic vacant spots and 50%haemolysis requirement for complement(HC_(50)).Immune group II:phagocytosis of phagocytic chicken erythrocytes in mice peritoneal macrophages and expression of CD4 and CD8 in spleen.[Results]Compared with the normal control group,all three doses of modified Duanggui Buxue Decoction significantly elevated the number of voluntary activities and grip strength,spleen index and thymus index of mice(P<0.05,P<0.01);enhanced the proliferative capacity of splenic lymphocytes and the activity of NK cells(P<0.01);increased the number of hemolytic vacant spots,HCso,the phagocytosis rate of chicken erythrocytes and phagocytosis index of the peritoneal macrophage(P<0.01);elevated whole blood lymphocyte(LYMPH),white blood cell(WBC)and red blood cell(RBC)counts,and serum IgM and IgG content(P<0.05,P<0.01);and decreased the proportions of CD3^(+)T and CD8^(+)T cells in blood and spleen,and decreased the amount of CD8 positive expression(P<0.05,P<0.01).Elevated peripheral blood CD19+B cell percentage,peripheral blood and spleen CD4+/CD8^(+)ratio and spleen CD4+/CD8^(+) positive expression ratio(P<0.05,P<0.01).[Conclusion]The modified Duanggui Buxue
