摘  要:目的:建立苗药紫藤根药材超高效液相色谱法(UPLC)指纹图谱及4个黄酮类成分含量测定方法,为全面评价其质量提供依据。方法:采用UPLC建立紫藤根药材指纹图谱,对不同产地药材进行相似度评价、聚类分析和主成分分析;同时对4个黄酮类成分含量进行测定。结果:建立了紫藤根UPLC指纹图谱,确定19个共有峰,指认了其中4个成分,分别为鸢尾苷、芒柄花苷、紫藤苷、biochanin A 7-O-glucoside。12批紫藤根药材相似度介于0.839至0.975之间;聚类分析可将不同产地的药材分为3类,表明不同产地药材存在一定差异;含量测定结果显示,安徽亳州产地药材鸢尾苷含量最高、湖南湘阴产地药材芒柄花苷含量最高,两地4个指标性成分总含量较高且稳定。结论:该方法可以有效评价不同产区紫藤根药材的质量差异性,为其质量控制和药材选购提供参考。Objective:To establish an ultra high performance liquid chromatography(UPLC)fingerprint and a method for determining the content of four flavonoid components in the seedling medicine Ziteng root,in order to provide a basis for comprehensive evaluation of its quality.Method:UPLC was used to establish a fingerprint of wisteria root,and similarity evaluation,cluster analysis,and principal component analysis were performed on different medicinal materials from different regions;Simultaneously measure the content of four flavonoids.Result:A UPLC fingerprint of wisteria root was established,19 common peaks were identified,and 5 components were identified,including iriside,anthocyanin,wisteria glycoside,and biochannin A7-O-glucoside.The similarity of 12 batches of wisteria root medicinal materials ranges from 0.839 to 0.975;Cluster analysis can classify medicinal materials from different regions into three categories,indicating that there are certain differences in medicinal materials from different regions;The content determination results showed that the content of iris glycoside in the medicinal herb from Bozhou,Anhui Province was the highest,while the content of anthocyanin in the medicinal herb from Xiangyin,Hunan Province was the best.The total content of the four indicator components in the two regions was relatively high and stable.Conclusion:This method can effectively evaluate the quality differences of wisteria root medicinal materials from different production areas,providing reference for quality control and medicinal material selection.
