摘  要:以树番茄带腋芽茎段为外植体,研究杀菌剂对外植体消毒及生长激素对愈伤组织、不定芽及不定根诱导的影响,同时探寻合适的组培苗移栽基质,为构建高效的树番茄无菌培养及植株再生体系提供参考。结果表明:用0.1%HgCl2消毒外植体15 min,成活率达97.8%;愈伤组织诱导最适宜激素配比为6-BA 0.4 mg/L+NAA 0.1 mg/L,诱导率达100%,不定芽诱导最适宜激素配比为6-BA0.6 mg/L+NAA 0.2 mg/L,不定芽诱导率达92.5%,平均分化芽数达到3个/块;不定根诱导最适宜激素配比为NAA 0.5 mg/L+IAA 0.1 mg/L,生根率达100%,平均生根数达4.2条,平均根长2.9 cm。炼苗7 d后移栽于腐殖土∶育苗基质∶椰糠(体积比2∶2∶1)的混合基质中,成活率达96%。The stem segment of tree tomato with axillary bud was used as explant to study the effects of fungicide disinfection and growth hormone on callus,adventitious bud and adventitious root induction,and to explore the suitable transplanting substrate for tissue culture plantlets.to provide reference for the construction of efficient aseptic culture and plant regeneration system of tree tomato.The results showed that the survival rate of explants sterilized with 0.1%HgCl2 for 15 minutes was 97.8%.The optimal hormone combination for callus induction was6-BA 0.4 mg/L+NAA 0.1 mg/L,and the induction rate was 100%.The optimum hormone combination for adventitious bud induction is 6-BA 0.6mg/L+NAA 0.2mg/L,and adventitious bud induction rate reached 92.5%,the average number of differentiated buds reached 3 per block.The optimal hormone combination for rooting was IAA 0.1 mg/L+NAA 0.5 mg/L,and the rooting rate was 100%,the average rooting number was 4.2,and the average root length was 2.9 cm.After 7 days of hardening-seedling,the survival rate was up to 96%when the mixture of seedling medium∶seedling medium∶coconut husk volume ratio of 2∶2∶1 was transplanted.
