摘  要:目的通过动物实验观察肉苁蓉干膏粉是否有致畸作用,初步确定不引起致畸作用的NOAEL值,为安全食用和相关产品开发提供参考。方法雌雄SD大鼠按1︰2合笼,查见阴栓当天为妊娠第0天,此后1 d为第1天。完全随机法将交配成功的雌鼠分入阴性对照组、1250 mg/kg、2500 mg/kg、5000 mg/kg、阳性对照组(环磷酰胺,15 mg/kg)5组,每组不少于18只。孕第6~15 d,剂量组以10 ml/kg经口灌胃给予受试物、阴性对照组灌胃给予相同体积蒸馏水、阳性组于孕第12天按2 ml/kg体重腹腔注射环磷酰胺1次。第0、6、9、12、15、20天称量体重,调整灌胃量。妊娠第20天,孕鼠称重后行安乐死,取出子宫,称子宫连胎鼠总重,摘除卵巢称重并计数黄体数,剖开子宫记录总着床数、活胎数、死胎数、吸收胎数,取出活胎仔称重、量体长、逐个检查外观,取下胎盘称重。将1/2活胎鼠放入鲍氏液中固定两周,检查脏器发育情况;其余活胎鼠去内脏,经固定、染色、透明后以检查骨骼发育情况。结果环磷酰胺组孕鼠增重显著低于阴性对照组(P=0.000),死胎发生率显著高于阴性对照组(P=0.000),胎仔平均体重及平均身长均显著低于阴性对照组(P=0.000、0.000),外观畸形、骨骼畸形及内脏畸形窝发生率均显著高于阴性对照组(χ^(2)=38.000、10286、18.000,P=0.000、0.001、0.000),提示试验系统及操作可靠。第0天(χ^(2)=5.664,P=0.227)、6天(F=0.906,P=0.464)、9天(F=1.098,P=0.363)、12天(F=1.081,P=0.371)、15天(F=0.627,P=0.644)孕鼠体重和体重净增重比较,组间差异无统计学意义(χ^(2)=5.790,P=0.215)。各剂量组第20天体重及体重增重与阴性对照组比较,差异无统计学意义(P=1.000、1.000、1.000,0.670、0.518、0.340)。窝平均活胎数(χ^(2)=1.869,P=0.760)、黄体数(χ^(2)=6.230,P=0.183)、吸收胎发生率(χ^(2)=1.109,P=0.894)组间差异无统计学意义;着床数(χ^(2)=9.964,P=0.041)、胎仔平均体重(χ^(2)=48.968,P=0Objective To observe the teratogenic effect of cistanche deserticola dried paste powder via animal experiment,and preliminarily confirm the dose that cistanche deserticola dried paste powder could not induce teratogenic effect(the NOAEL value),and provide a reference for edible safety and related product development.Methods Female and male SD rats were mated at a ratio of 1︰2.The day the vaginal plug was found was defined as the day 0 of pregnancy,and the next day was defined as the day 1 of pregnancy.The successfully mated female rats were divided into five groups of negative control group,1250 mg/kg,2500 mg/kg,5000 mg/kg,and positive control group(cyclophosphamide,15 mg/kg)according to a completely random method,finally no less than 18 females in each group.From the 6th to 15th day of pregnancy,the rats in each dose group were given the test substance(10 ml/kg)by gavage,and the negative control group was given the same volume of distilled water by gavage,while on the 12th day of pregnancy the rats in the positive control group were intraperitoneal injected with cyclophosphamide at the volume of 2 ml/kg.On day 0,6,9,12,15,and 20,the body weight of pregnant rats were measured and the gavage volume was adjusted.On the 20th day of pregnancy,the pregnant rats were weighed and euthanized;the uterus was removed,the total weight of the uteruses which enfolded fetal rats were measured;the ovaries were removed and weighed;the number of corpus luteum were counted;the total number of embryo implantation,number of live fetuses,number of dead fetuses,number of absorbed fetuses were recorded;the weight and length of the live fetuses were measured,and the placentas were removed and weighed.Half of the live fetal rats were dropped into bowman's solution and fixed for two weeks,and then the development situation of organs in rat fetuses were inspected,while the rest of the live fetal rats were euthanized,and internal organs in chest and abdominal cavity were removed,after the process of fixation,dyeing,and transparent,bone dev
