摘 要:以黄芪为试材,利用水提醇沉法提取多糖,采用正交试验考查乙醇浓度、提取时间、提取温度、料液比4个试验因素,以黄芪多糖提取率为试验指标进行工艺条件优化。结果表明:黄芪多糖提取工艺为提取时间1.5 h、提取温度100℃、料液比1∶6 g/mL、90%的乙醇隔夜沉淀。在此条件下,黄芪多糖提取率最高,为2.85%。通过正交试验设计优化黄芪多糖提取工艺稳定可行,为黄芪多糖提取方法的研究提供参考。Astragalus was used as test material.Polysaccharides were extracted by water boiling and alcohol precipitation method.Orthogonal test was used to investigate the factors of ethanol concentration,extraction time,extraction temperature,and the ratio of material to liquid.The extraction rate of Astragalus polysaccharide was used as the test index to optimize the process conditions.The results showed that the extraction process of Astragalus polysaccharide was the extraction time 1.5 hours,the extraction temperature 100℃,theratio of material to liquid 1:6g/mL,and 90%alcohol was precipitated overnight.Under this condition,the extraction rate of Astragalus polysaccharide was the highest,which was 2.85%.It is stable and feasible to optimize the extraction process of Astragalus polysaccharide by orthogonal test design,which provides a reference for the study of Astragalus polysaccharide extraction method.
